Tumor-penetrating therapy for β5 integrin-rich pancreas cancer

Summary: CAFs induce beta 5 integrin expression in tumor cells in a TGF-beta dependent manner, making them an efficient drug delivery target for the tumor-penetrating peptide iRGD. Knocking out beta 5 integrins in tumor cells suppresses the capacity of iRGD to deliver payloads, leading to reduced disease burden and prolonged survival in mouse models. iRGD penetration into the desmoplastic PDAC microenvironment requires beta 5 integrin expression on tumor cells mediated by CAFs-derived TGF-beta, and iRGD-based combination therapy is effective in PDAC mouse models.

TGF-β Signaling Promotes Glioma Progression Through Stabilizing Sox9

Summary: Sox9, an upregulated transcription factor in clinical gliomas associated with poor prognosis, promotes migration and invasion of glioma cells and in vivo tumor development. It functions downstream of the TGF-beta pathway, where TGF-beta signaling prevents proteasomal degradation of Sox9 protein in glioma cells. These findings provide new insights into the interaction between TGF-beta signaling and oncogenic transcription factors, with implications for targeted therapy and prognostic assessment of gliomas.

Summary: The study revealed a correlation between high CTSS expression and glioma tissue grades, with CTSS inhibitor ZFL attenuating TGF-beta-induced invasive growth. Inhibition of CTSS reversed TGF-beta-induced epithelial-to-mesenchymal transition and restored the turnover of tight junction proteins, leading to decreased mobility of glioblastoma cells. Furthermore, the PI3K/AKT/mTOR pathway was found to be suppressed in the TGF-beta+ZFL groups, highlighting its importance in this process.
