Rethinking organoid technology through bioengineering

Elena Garreta et al.

Summary: Significant progress has been made in the differentiation of human pluripotent stem cells and the development of organoids. The convergence of stem cell biology and bioengineering allows for controlled stimuli to be provided to the system. Engineering solutions are emphasized to control the self-organization and functionality of stem cell-derived organoids.

Organoids and organ chips in ophthalmology

Navid Manafi et al.

Summary: Recent advances in stem cell-derived, self-organizing, three-dimensional miniature organs, such as organoids and organ chips, have shown potential in simulating the complexity of native organ structure and function. They may enhance our understanding of the physiology of different eye structures, explore unknown aspects of ophthalmic disease pathogenesis, and serve as clinically-relevant tools for evaluating ocular therapeutics.

Harmonization of Protocols for Multi-Species Organoid Platforms to Study the Intestinal Biology of Toxoplasma gondii and Other Protozoan Infections

David Holthaus et al.

Summary: Utilizing an in vitro platform with stem cell-derived intestinal organoids, models for human, mouse, pig, and chicken were established for studying mechanisms of infection by intestinal protozoan parasites. The generation, differentiation, and characterization of these organoids were optimized, leading to successful applications in comparative studies on parasite-host interactions and co-infections with relevant protozoans.

Inside-out chicken enteroids with leukocyte component as a model to study host-pathogen interactions

Tessa J. Nash et al.

Summary: Researchers have developed complex 3D chicken enteroids that mimic intestinal organization, allowing for investigation of intestinal cell biology and host-pathogen interactions. These enteroids can develop optimally in suspension and have successfully been infected with Salmonella Typhimurium, influenza A virus, and Eimeria tenella without the need for micro-injection, providing a comprehensive model for exploring epithelial and leukocyte interactions in host-pathogen, food science, and pharmaceutical research.

Plasmodium vivax Gametocytes Adherence to Bone Marrow Endothelial Cells

Luis Carlos Salazar Alvarez et al.

Summary: In a Plasmodium vivax infection, gametocytes show increased distribution in bone marrow aspirant, indicating the organ's role as a parasite reservoir. Ex vivo assays demonstrate gametocyte adherence to bone marrow endothelial cells, with ICAM1 as a significant receptor and CD36 showing high adhesion rate without significance.

Toxoplasma gondii excreted/secreted proteases disrupt intercellular junction proteins in epithelial cell monolayers to facilitate tachyzoites paracellular migration

Carlos J. Ramirez-Flores et al.

Summary: The study reveals that the proteases present in Toxoplasma ESPs disrupt intercellular junctions of epithelial monolayers, facilitating paracellular migration of the parasite. Pre-treatment with protease inhibitors prevents the disruption of tight junction proteins and loss of transepithelial electrical resistance induced by ESPs. The internalization of TJ proteins is triggered by ESPs proteases, leading to the observed modifications in cell junctions.
