High-throughput rational design of the remdesivir binding site in the RdRp of SARS-CoV-2: implications for potential resistance

Summary: With the use of mutational mapping, it was predicted that only a few mutations in the nsp12 subunit of RdRp are needed for SARS-CoV-2 to develop resistance to remdesivir. Certain mutations resulted in decreased binding affinity to remdesivir, indicating drug resistance. Identifying potential hotspot residues prone to mutation improves our understanding of SARS-CoV-2 drug resistance and COVID-19 pathogenesis.

Site-specific N-glycosylation analysis of animal cell culture-derived Zika virus proteins

Summary: This study presents a site-specific N-glycosylation analysis of proteins from a Brazilian Zika virus (ZIKV) strain for the first time, revealing high-mannose glycosylation on NS1 protein and highly heterogeneous N-glycosylation on E protein. The established method shows promise for characterizing viral isolates, monitoring glycosylation of major antigens, and can be applied to study the impact of protein glycosylation on antigenicity and human pathogenicity of various viruses.
