Accuracy of cholera rapid diagnostic tests: a systematic review and meta-analysis

Basilua Andre Muzembo et al.

Summary: This study systematically evaluated the accuracy of cholera rapid diagnostic tests (RDTs) and found that they have moderate sensitivity and specificity, especially when enriched with alkaline peptone water (APW). Although they are less useful for clinical management, the current generation of RDTs can be valuable for surveillance efforts if used in a principled manner.

Predicting Vibrio cholerae Infection and Disease Severity Using Metagenomics in a Prospective Cohort Study

Ines Levade et al.

Summary: This study utilized deep shotgun metagenomic sequencing to analyze household contacts of cholera patients, successfully identifying species, strains, gene families, and cellular pathways in the gut microbiome associated with predicting cholera infection and symptoms. Additionally, specific species and genes were highlighted as potential targets for experimental testing to investigate microbiome-related protection mechanisms against cholera.

Highly targeted spatiotemporal interventions against cholera epidemics, 2000-19: a scoping review

Ruwan Ratnayake et al.

Summary: Cholera epidemics continue to pose challenges to disease control globally. Targeted interventions such as antibiotic chemoprophylaxis, point-of-use water treatment, and hygiene promotion have been found effective in rapidly reducing household transmission. Early detection capacity is critical for the success of case-area targeted interventions for outbreak control.

A multifaceted cellular damage repair and prevention pathway promotes high-level tolerance to β-lactam antibiotics

Jung-Ho Shin et al.

Summary: Bactericidal antibiotics are powerful drugs that can convert essential bacterial functions into lethal processes, but some bacterial pathogens are remarkably tolerant to these antibiotics. The VxrAB system of Vibrio cholerae contributes to high-level beta-lactam tolerance by controlling gene networks that regulate iron uptake and metabolism. Sequestration of iron enhances tolerance against beta-lactam therapy in a mouse model of cholera infection.

Laboratory and Field Evaluation of the Crystal VC-O1 Cholera Rapid Diagnostic Test

Summary: Cholera is a severe diarrheal disease that affects many low-and middle-income countries. Current rapid diagnostic tests primarily target serotype O1, demonstrating high sensitivity and specificity, which could significantly improve detection and surveillance of cholera outbreaks.

Impact of Immunoglobulin Isotype and Epitope on the Functional Properties of Vibrio cholerae O-Specific Polysaccharide-Specific Monoclonal Antibodies

Robert C. Kauffman et al.

Summary: The properties and protective mechanisms of functionally relevant antibodies against Vibrio cholerae have been investigated, showing that affinity, immunoglobulin isotype, and subclass do not directly dictate functional potency. Importantly, low-affinity variants displayed comparable agglutination and motility inhibition properties to high-affinity antibodies, suggesting that the way the antibody binds to the O-antigen may be critical to function.

Serum vibriocidal responses when second doses of oral cholera vaccine are delayed 6 months in Zambia

John Mwaba et al.

Summary: Two-dose killed oral cholera vaccines (OCV) are widely used for cholera control. This study compared the vibriocidal response induced by OCV with standard dose-interval (2 weeks) and extended dose-interval (6 months) for the second dose. The findings indicated that vibriocidal titers 14 days after the second dose were similar between the standard and extended dose-interval groups.

Toward Cholera Elimination, Haiti

Stanislas Rebaudet et al.

Summary: The study shows that there has been a significant decrease in cholera transmission in Haiti since February 2019, with case-area targeted interventions by rapid response teams playing a key role. However, vulnerabilities persist and vaccination remains limited, leading to questions about the presence of environmental reservoirs in Haiti and progress toward elimination.
