Multiplexed Ion Beam Imaging: Insights into Pathobiology

Candace C. Liu et al.

Summary: Next-generation tools for multiplexed imaging have revolutionized our understanding of single-cell function and tissue structure. By using multiplexed ion beam imaging, we can visualize multiple antibodies tagged with metal reporters, increasing the breadth and depth of imaging data. To analyze these data effectively, we have developed tools for cell identification, cell classification, and spatial analysis. These tools have been applied in various disease contexts and hold great potential for improving diagnosis, prognosis, and therapeutic selection.

Transition to invasive breast cancer is associated with progressive changes in the structure and composition of tumor stroma

Tyler Risom et al.

Summary: This study used MIBI-TOF and antibody staining methods to analyze the changes in the tumor microenvironment (TME) between DCIS and IBC. Four coordinated transitions between TME states were identified. The study also found that myoepithelial disruption was more advanced in DCIS patients who did not develop IBC, suggesting a potential protective effect against recurrence.

Network for Biomarker Immunoprofiling for Cancer Immunotherapy: Cancer Immune Monitoring and Analysis Centers and Cancer Immunologic Data Commons (CIMAC-CIDC)

Helen X. Chen et al.

Summary: Immunoprofiling for identifying biomarkers and integrating with clinical trial outcomes play a critical role in improving immunotherapy for cancer patients. The CIMAC-CIDC Network aims to establish an infrastructure for systematic biomarker identification and correlation with clinical outcomes across trials, facilitated by harmonized assays and computational expertise. By establishing this network, immuno-oncology biomarker development and cross-trial data analysis can be enhanced.

MAUI (MBI Analysis User Interface)-An image processing pipeline for Multiplexed Mass Based Imaging

Alex Baranski et al.

Summary: MAUI is a series of graphical user interfaces designed to streamline data preprocessing steps, including the removal of crosstalk, noise, and antibody aggregates, to accelerate and automate processing. It enables real-time and interactive parameter tuning, providing high-quality and reproducible data input for downstream analyses.
