Targeting cellular senescence with senotherapeutics: senolytics and senomorphics

Summary: The concept of geroscience focuses on targeting the ageing process itself as a means to improve human health. Cellular senescence, a state of irreversible cell arrest, has emerged as a potential therapeutic target to extend healthspan. Senescent cells develop a secretory phenotype that promotes chronic inflammation and tissue dysfunction. Therapeutic drugs that induce selective cell death of senescent cells or suppress the expression of senescence markers are being developed.

The senescence of lung epithelial cells and regulation of miRNAs have been found to be associated with COPD development.

miR-146a impedes the anti-aging effect of AMPK via NAMPT suppression and NAD+/SIRT inactivation

Summary: In this study, researchers found that miR-146a negatively regulates the AMPK and NAD(+)/SIRT pathway by inhibiting the expression of NAMPT. Additionally, AMPK activators suppress the expression of miR-146a, further enhancing NAMPT expression. This mutual inhibitory relationship provides new insights into the molecular connections between AMPK and SIRT and offers a potential intervention point for the prevention of aging and age-related diseases.

Cellular senescence in ageing: from mechanisms to therapeutic opportunities

Apart from stress-induced senescence, it can also be part of controlled programs in biological processes like embryonic development. Senescent cells contribute to impaired tissue regeneration, chronic diseases, and organismal aging, but also present an opportunity for intervention in aging and associated diseases.

p21 produces a bioactive secretome that places stressed cells under immunosurveillance

Summary: The cell-cycle inhibitor p21 activates Rb-dependent transcription to generate a complex bioactive secretome, known as PASP, which includes chemokine CXCL14 that attracts macrophages. Macrophages disengage if cells normalize p21 within 4 days, but persisting p21 induction leads to M1 polarization and cytotoxic T cell response for eliminating target cells, including preneoplastic cells. Thus, p21 concurrently induces proliferative arrest and immunosurveillance of cells under duress.

Oxylipin biosynthesis reinforces cellular senescence and allows detection of senolysis

Summary: Cellular senescence is a stress or damage response that leads to permanent proliferative arrest and secretion of factors with potent biological activities. Senescent cells activate the biosynthesis of oxylipins, including prostaglandins, which promote the senescence arrest and SASP by activating RAS signaling. The release of specific prostaglandins, such as 15deoxy-delta-12,14-prostaglandin J2, serves as a biomarker for senolysis in culture and in vivo.
