The impact of storage on extracellular vesicles: A systematic study

Stefano Gelibter et al.

Summary: Storage has a significant impact on extracellular vesicles (EVs) samples, leading to a decrease in concentration, reduction in purity, and generation of artefactual particles. It is recommended to process fresh, non-archival samples for EVs analysis, depending on downstream analyses and experimental settings.

Factor VIIa suppresses inflammation and barrier disruption through the release of EEVs and transfer of microRNA 10a

Kaushik Das et al.

Summary: This study reveals the interaction mechanism between FVIIa and endothelial cells, as well as the anti-inflammatory and vascular protective effects of FVIIa-released EVs. The results show that FVIIa induces the release of EEVs from endothelial cells through activation of PAR1 and the ROCK-dependent pathway. These EEVs are enriched with anti-inflammatory microRNAs, primarily miR10a. EEVs can be taken up by monocytes/macrophages and endothelial cells, thereby modulating their anti-inflammatory and barrier protective effects.

Factor VIIa induces extracellular vesicles from the endothelium: a potential mechanism for its hemostatic effect

Summary: Studies showed that FVIIa induces the release of EVs from endothelial cells through the EPCR-PAR1 signaling pathway, contributing to the hemostatic effect of FVIIa.

Summary: The study summarized evidence from three different sources demonstrating the efficacy and safety of rFVIIa in patients with GT without refractoriness or antibodies to platelets, supporting the label extension of rFVIIa as an alternative treatment when platelets are unavailable.

Preserving extracellular vesicles for biomedical applications: consideration of storage stability before and after isolation

Fumin Yuan et al.

Summary: Extracellular vesicles (EVs) are nanovesicles released by various cell types, known for their potent biological activities. While -80 degrees C is considered as the most favorable condition for storage, challenges remain in understanding the impact of storage conditions on EVs, indicating further research is needed.
