Partial purification, characterization and kinetics of thermal inactivation of pectin methylesterase and polygalacturonase enzymes from Indian lemon (Citrus limon (L.)

Summary: The extraction, purification, and characterization of PME and PG enzymes from lemon fruit were successfully carried out using ion exchange chromatography and SDS-PAGE analysis. The optimal working conditions and activation energies of PME and PG enzymes were determined. The stability of the enzymes was not significantly affected by the fruit matrix, providing insights for designing thermal treatments.

Summary: Proteins acting as potent inhibitors of plant pectin methylesterase and polygalacturonase were isolated and purified from lemon fruits. The inhibitors exhibited stability and optimal pH and temperature conditions, with the polygalacturonase inhibitor protein being more heat stable. The inhibition kinetics of the inhibitors were noncompetitive, making them potential alternatives to thermal processing in fruit-based products.
