Summary: This review provides an overview of the available patent literature on JAK-family inhibitors and the clinical activities of selective JAK3 inhibitors. Although no selective JAK3 inhibitors have been approved for clinical use, recent studies have shown the promising therapeutic potential of these inhibitors in inflammatory and autoimmune diseases.

Ritlecitinib and brepocitinib demonstrate significant improvement in scalp alopecia areata biomarkers

Summary: JAK inhibitors (ritelcitinib and brepocitinib) have shown promising results in the treatment of alopecia areata. Both drugs demonstrated improvements in the gene expression of scalp tissue, with ritlecitinib showing better results in the later stages of treatment. The improvement in the Severity of Alopecia Tool (SALT) scores was positively associated with the expression of T(H)1 markers and negatively associated with the expression of hair keratins.

Characterizing the relationships between patient-reported outcomes and clinician assessments of alopecia areata in a phase 2a randomized trial of ritlecitinib and brepocitinib

Summary: This study evaluated the efficacy and impact of ritlecitinib and brepocitinib on patients with alopecia areata through the ALLEGRO trial. The patient-reported outcome tool AASIS showed correlation with clinician-assessed SALT scores, and both ritlecitinib and brepocitinib groups had significant improvements in symptom and severity scores at week 24 compared to placebo.
