Mitochondrion targeting peptide-modified magnetic graphene oxide delivering mitoxantrone for impairment of tumor mitochondrial functions

Hangqi Zhu et al.

Summary: This study achieved efficient impairment of tumor mitochondria by preparing mitochondrion targeting peptide-grafted magnetic graphene oxide nanocarriers. The nanocarriers exhibited high drug-loading capacity for anticancer drug delivery and preferential targeting of tumor mitochondria, leading to severe impairment of mitochondrial functions and activation of apoptosis. The study demonstrates the potential of two-dimensional nanoplatforms for mitochondrion-targeting drug delivery in cancer therapy.

PEG-GO@XN nanocomposite suppresses breast cancer metastasis via inhibition of mitochondrial oxidative phosphorylation and blockade of epithelial-to-mesenchymal transition

Jialing Zhang et al.

Summary: In this study, a graphene-based nanocomposite PEG-GO@XN was developed to selectively inhibit mitochondrial oxidative phosphorylation in metastatic breast cancer cells, effectively blocking their migration and invasion. Additionally, the composite suppressed epithelial-mesenchymal transition in cancer cells and reduced their motility potential, offering a promising approach for metastatic breast cancer treatment.

Spatiotemporally programmable cascade hybridization of hairpin DNA in polymeric nanoframework for precise siRNA delivery

Feng Li et al.

Summary: DNA nanostructures have been used as carriers for gene delivery, but spatiotemporally programmable assembly of DNA under nanoconfinement is challenging due to DNA complexity. A strategy using cascade hybridization chain reaction of DNA hairpins in polymeric nanoframework enables precise siRNA delivery. The nanoframework provides stability and ease of functionalization, addressing DNA complexity issues. Phenylboronate installation on nanoframework enhances cellular uptake and lysosomal escape.

Supramolecular Antagonists Promote Mitochondrial Dysfunction

Mingming Li et al.

Summary: Mitochondrion-targeting therapy has great potential in cancer treatment, but limited therapeutic efficiency is a challenge. A mitochondrion-targeting supramolecular antagonist was developed, inducing tumor cell death by promoting apoptosis and preventing survival simultaneously. The combined therapeutic effect of the BH3 domain and CPT drug in the supramolecular antagonist efficiently inhibits tumor growth.

Cell-penetrating peptide-conjugated lipid/polymer hybrid nanovesicles for endoplasmic reticulum-targeting intracellular delivery

Jeong Yi Kang et al.

Summary: The study presents an intracellular delivery approach utilizing cell-penetrating peptide (CPP)-conjugated lipid/polymer hybrid nanovehicles (LPNVs) for ER-targeting, showing efficient uptake by cells and specific ER-targeting ability. Remarkable lysosomal escape of the LPNVs was observed due to effective pH buffering with the aid of PEO-b-PCL-b-PEO, indicating the potential of the LPNVPnt system for elaborate drug delivery technology development at the intracellular level.
