Biocatalytic One-Carbon Ring Expansion of Aziridines to Azetidines via a Highly Enantioselective [1,2]-Stevens Rearrangement

Copper-Catalyzed Asymmetric Diyne Cyclization via [1,2]-Stevens-Type Rearrangement for the Synthesis of Chiral Chromeno[3,4-c]pyrroles

Summary: In this study, a copper-catalyzed asymmetric cascade cyclization/[1,2]-Stevens-type rearrangement was reported to efficiently assemble a variety of valuable chiral chromeno[3,4-c]pyrroles bearing a quaternary carbon stereocenter with generally moderate to good yields and excellent enantioselectivities (up to 99% ee). Notably, this protocol represents the first example of catalytic asymmetric [1,2]-Stevens-type rearrangement based on alkynes and the first asymmetric formal carbene insertion into the Si-O bond.

Tunable Aziridinium Ylide Reactivity: Noncovalent Interactions Enable Divergent Product Outcomes

Summary: This work describes the methods for rapidly preparing densely functionalized and stereochemically complex N-heterocyclic scaffolds. Experimental and computational studies were conducted to better understand the features of aziridinium ylides as intermediates in the synthesis of highly substituted dehydromorpholines. Several key insights were uncovered, including the importance of steric effects, rotational barriers, and noncovalent interactions. These insights will contribute to the development of this chemistry for expanding the complex amine chemical space.
