Quantifying the DNA-damaging Effects of FLASH Irradiation With Plasmid DNA

Alan Perstin et al.

Summary: This study investigated the DNA-damaging effects of ultrahigh-dose-rate (FLASH) irradiation compared to conventional dose-rate irradiation using a plasmid DNA nicking assay. The results showed that FLASH irradiation caused less damage to DNA compared to the conventional dose rate, supporting the idea that the protective effect of FLASH irradiation occurs at least partially through fundamental biochemical processes.

Profiling DNA Damage Induced by the Irradiation of DNA with Gold Nanoparticles

Alaa Huwaidi et al.

Summary: The presence of gold nanoparticles enhances the formation of DNA damage under therapeutic X-ray exposure. The major damage includes the release of nonmodified nucleobases and the formation of common reduction and oxidation products of DNA, as well as unique modifications of the 2-deoxyribose moiety. Most of the damage is believed to be caused by the generation of Auger/low-energy electrons and their reaction with DNA.

Evaluating very high energy electron RBE from nanodosimetric pBR322 plasmid DNA damage

K. L. Small et al.

Summary: This paper presents the first plasmid DNA irradiations carried out with Very High Energy Electrons (VHEE) over 100-200 MeV at the CLEAR user facility at CERN to determine the Relative Biological Effectiveness (RBE) of VHEE. The study found similarities in DNA damage yields between VHEE and conventional modalities, and reasonable agreement between experimental values and current models for DSB yield. Additionally, no significant variations in DSB yield with dose rate were observed following VHEE irradiation, indicating no FLASH effect under these conditions.

Folding DNA into origami nanostructures enhances resistance to ionizing radiation

Leo Sala et al.

Summary: DNA origami triangles exhibit extreme stability upon irradiation, attributed to the multi-row design and protective effect of the folded structure. The protective effect of this structure surpasses that of a naturally paired DNA double helix, opening up possibilities for the use of scaffolded DNA in designing radiation-resistant and radio-sensitive materials.

DNA origami

Swarup Dey et al.

Summary: DNA origami technology, a bottom-up method for constructing nanostructures, offers high precision and controllability. It finds wide applications in nanofabrication, nanophotonics, nanoelectronics, catalysis, computation, bioimaging, and more. However, challenges such as size limits, stability issues, and scale of production need to be addressed for further advancement in the field.
