Applications of Halogen-Atom Transfer (XAT) for the Generation of Carbon Radicals in Synthetic Photochemistry and Photocatalysis

Fabio Julia et al.

Summary: This review focuses on the importance and application of halogen-atom transfer (XAT) in synthetic chemistry, particularly its impact on photochemistry and photocatalysis. The material is organized based on mechanistic aspects and the nature of reagents used in halogen abstraction. The aim is to provide a general overview of fundamental concepts and key agents involved in XAT processes to facilitate the development of new synthetic radical strategies.

Access to gem-Difluoroalkenes via Organic Photoredox-Catalyzed gem-Difluoroallylation of Alkyl Iodides

Songlin Yan et al.

Summary: An organic photoredox-catalyzed gem-difluoroallylation of alpha-trifluoromethyl alkenes with alkyl iodides via C-F bond cleavage for the synthesis of gem-difluoroalkene derivatives is reported. The transition-metal-free transformation utilized a readily available organic dye as the sole photocatalyst and provided good to high yields with a variety of iodides, including primary, secondary, and tertiary alkyl iodides.

Ni/Photoredox-Catalyzed Enantioselective Cross-Electrophile Coupling of Styrene Oxides with Aryl Iodides

Sii Hong Lau et al.

Summary: This study introduces a Ni/photoredox-catalyzed enantioselective reductive coupling of styrene oxides and aryl iodides, yielding enantioenriched 2,2-diarylalcohols. Analysis shows that enantioselectivity is correlated with the electronic properties of the ligands, with more electron-donating ligands leading to higher ee values. Experimental and computational studies support the hypothesis that reductive elimination is enantiodetermining and ligand electronic properties affect enantioselectivity by altering the position of the transition state structure.

Visible-Light-Mediated C-I Difluoroallylation with an α-Aminoalkyl Radical as a Mediator

Fuyang Yue et al.

Summary: This protocol involves direct visible-light-mediated C-I difluoroallylation reactions of α-trifluoromethyl arylalkenes with alkyl iodides at room temperature using an alpha-aminoalkyl radical as a mediator. It allows efficient functionalization of various alpha-trifluoromethyl arylalkenes with cyclic and acyclic primary, secondary, and tertiary alkyl iodides and is scalable to gram level. The mild protocol using an inexpensive mediator is suitable for late-stage functionalization of complex natural products and drugs.

A general N-alkylation platform via copper metallaphotoredox and silyl radical activation of alkyl halides

Nathan W. Dow et al.

Summary: A catalytic union of various functional groups with electronically and sterically diverse alkyl bromides has been achieved through a visible-light-induced metalla-photoredox platform, bypassing traditional high barriers. This regio- and chemoselective protocol is compatible with a wide range of medicinally relevant N-nucleophiles and structurally diverse alkyl bromides. Preliminary mechanistic experiments validate the dual catalytic, open-shell nature of this platform, enabling reactivity previously unattainable in traditional halide-based N-alkylation systems.

Summary: The study demonstrates an alternative amination strategy using alkyl iodides as radical precursors and copper catalysis to assemble sp3 C-N bonds at room temperature. This method provides S(N)2-like programmability and has been shown to be useful in the late-stage functionalization of pharmaceuticals for the preparation of valuable N-alkylated drug analogues.

Phenolate anion-catalyzed direct activation of inert alkyl chlorides driven by visible light

Delian Wei et al.

Summary: This study demonstrates an efficient photochemical strategy to activate inert C(sp(3))-Cl bonds using phenolate anion photocatalysts. Mechanistic studies show that the cleavage of unactivated C(sp(3))-Cl bonds is mediated by a direct SET process from photo-excited phenolate anions. This strategy allows for the engagement of a variety of unactivated alkyl chlorides in radical C-O bond formation, dehalogenation, and cyclization under mild conditions.

Radical hydroxymethylation of alkyl iodides using formaldehyde as a C1 synthon

Lewis Caiger et al.

Summary: In this study, a method for hydroxymethylation using formaldehyde as a C1 synthon is achieved through photocatalysis and a phosphine additive. By leveraging halogen-atom transfer and capturing the O-radical, the addition reaction to formaldehyde is rendered irreversible, leading to improved efficiency and yield of the reaction.
