GLIAL CONNEXINS AND PANNEXINS IN THE HEALTHY AND DISEASED BRAIN

Christian Giaume et al.

Summary: Over the past few decades, research has shown that glial cells interact dynamically with neurons, impacting their activity and survival. Connexins and pannexins play a key role in these interactions, and understanding their function can provide insights into neuronal behavior and aid in the development of therapeutic strategies for brain diseases.

Connexin43 hemichannel block inhibits NLRP3 inflammasome activation in a human retinal explant model of diabetic retinopathy

Henry H. Louie et al.

Summary: Recent research has shown that connexin43 hemichannel blockers can inhibit activation of the inflammasome pathway in diabetic retinopathy. The development of an ex vivo human retinal organotypic culture model provides a more accurate representation of the inflammatory process in diabetic retinopathy, supporting the potential use of connexin43 hemichannel blockers as new therapies for the disease.

Connexin Gap Junctions and Hemichannels Link Oxidative Stress to Skeletal Physiology and Pathology

Rui Hua et al.

Summary: Oxidative stress plays a significant role in affecting the function of connexin channels and bone loss, with increased opening of connexin hemichannels under oxidative stress conditions providing cell protection. Additionally, oxidative stress decreases osteocyte viability and impairs communication among osteocytes.

Astrocyte dystrophy in ageing brain parallels impaired synaptic plasticity

Alexander Popov et al.

Summary: This study found a significant reduction in the number and length of astrocytic processes, astrocytic territorial domains, and astrocyte-to-astrocyte coupling in the aged brain. Physiological deficits in K+ and glutamate clearance, as well as spatiotemporal reorganisation of Ca2+ events, were observed in old astrocytes, which paralleled impaired synaptic long-term potentiation (LTP) in hippocampal CA1 in old mice. These findings may explain the astroglial mechanisms of age-dependent decline in learning and memory.

Orthopedic surgery-induced cognitive dysfunction is mediated by CX3CL1/R1 signaling

Inja Cho et al.

Summary: Postoperative pain is closely related to the development of postoperative cognitive dysfunction (POCD), and CX3CR1 signaling is identified as an important target for potential therapeutic strategies to prevent the development of POCD in a mouse model. The study found that increased levels of CX3CR1 after surgery were necessary and sufficient to induce cognitive dysfunction, highlighting the significance of targeting CX3CR1 in preventing POCD.

Isoflurane affects brain functional connectivity in rats 1 month after exposure

Petteri Stenroos et al.

Summary: Isoflurane anesthesia induces long-term changes in brain function and gene expression, leading to behavioral and cognitive consequences. Functional connectivity between thalamus-cortex and hippocampus-cortex was strengthened after isoflurane treatment, accompanied by altered expression of cortical genes involved in signal transmission. Further studies are needed to investigate the association of these changes with postoperative behavioral and cognitive symptoms.

Astrocytic processes: from tripartite synapses to the active milieu

Alexey Semyanov et al.

Summary: A new concept of 'active milieu' is defined in this study, which integrates all components of nervous tissue into a dynamic information processing system, with a focus on the role of astrocytic processes. These processes are involved in reciprocal interactions with neuronal compartments and communicate with other elements of the nervous tissue within this framework.

Amelioration of Hippocampal Insulin Resistance Reduces Tau Hyperphosphorylation and Cognitive Decline Induced by Isoflurane in Mice

Liangyu Peng et al.

Summary: Research shows that long-term exposure to isoflurane in mice can lead to hippocampal insulin resistance and tau hyperphosphorylation, resulting in cognitive impairment. Metformin can mitigate these effects by alleviating hippocampal insulin resistance and reversing tau hyperphosphorylation.

Astroglial Connexins in Neurodegenerative Diseases

Xiaomin Huang et al.

Summary: Astrocytes play a crucial role in maintaining the normal functions of the Central Nervous System (CNS). During neurodegenerative diseases, alterations in astrocytic connexin expression and function can affect disease progression by impacting neuronal function and survival. Targeting connexins may be a plausible therapeutic strategy for managing these neurodegenerative diseases.

Astrocyte Networks as Therapeutic Targets in Glaucomatous Neurodegeneration

Andrew M. Boal et al.

Summary: Astrocytes play a crucial role in responding to neurodegenerative stress and are interconnected through gap junctions, primarily composed of connexin-43 (Cx43), forming a broader network. This network contributes to various important processes and is likely to influence the progression of neurodegenerative diseases. Targeting these networks may be critical in the treatment of glaucomatous neurodegeneration and other optic neuropathies.
