Mapping the immune environment in clear cell renal carcinoma by single-cell genomics

Nicholas Borcherding et al.

Summary: The study utilized single-cell RNA sequencing to analyze the tumor immune microenvironment of ccRCC patients and found an increase in CD8(+) T cells and macrophages in tumor-infiltrated immune cells, as well as identified a proliferation subpopulation associated with the progression of ccRCC. Utilizing gene expression analysis, the study also demonstrated the ability to predict clinical outcomes and pathological diseases based on subcluster assignment.

Neurosarcoidosis Pathophysiology, Diagnosis, and Treatment

Michael J. Bradshaw et al.

Summary: Neurosarcoidosis typically presents as recognizable syndromes that can be diagnosed by considering clinical syndrome, imaging and histopathological findings, and excluding other causes. Recent years have seen significant advancements in understanding and treating neurosarcoidosis, with tumor necrosis factor inhibitors playing an important role in therapy.

Tofacitinib as a Steroid-Sparing Therapy in Pulmonary Sarcoidosis, an Open-Label Prospective Proof-of-Concept Study

Marcia A. Friedman et al.

Summary: In this proof-of-concept study, adding a JAK-inhibitor allowed 60% of patients with pulmonary sarcoidosis to successfully taper corticosteroids, showing potential as a promising therapy for this condition. Further investigation in randomized trials is warranted for JAK-inhibitors in the treatment of pulmonary sarcoidosis.

CD4+ T cells in the lungs of acute sarcoidosis patients recognize an Aspergillus nidulans epitope

Sarah A. Greaves et al.

Summary: The acute form of sarcoidosis known as Lofgren's syndrome is genetically associated with HLA-DR3 and characterized by an accumulation of CD4(+) T cells in bronchoalveolar lavage. Research has identified a peptide from Aspergillus nidulans that stimulates CD4(+) T cells in an HLA-DR3-restricted manner in LS patients. Additionally, LS subjects showed increased IFN-gamma and IL-2-secreting T cells in response to the NDPD peptide, along with elevated IgG antibody responses to A. nidulans NDPD in their serum. These findings suggest a potential role of A. nidulans in the etiology of LS.

Challenges of Sarcoidosis and Its Management

Marjolein Drent et al.

Summary: Sarcoidosis is a lung disease characterized by noncaseating granulomas of unknown cause, which can also affect the heart and nervous system. Tissue biopsy is required for definitive diagnosis, and glucocorticoids are recommended as initial treatment when necessary, but not for long-term care.

Single-cell transcriptome and TCR profiling reveal activated and expanded T cell populations in Parkinson's disease

Pingping Wang et al.

Summary: The study reveals the presence of a large population of CD8(+) T cells progressing continuously and remarkably expanded CD4 CTLs in PD patients through TCR tracking. Additionally, potential TCR-antigen pairs were identified, providing evidence for peripheral T cells to participate in neuronal degeneration.

Summary: The study identified unique molecular features in patients with granulomatous uveitis, showing distinct antigen-driven pathogenic processes. CD4 T cell populations exhibited individual variability, while ocular CD8 T cells and B cells were more transcriptionally similar among patients, with ocular B cells displaying evidence of class switching and plasmablast differentiation.
