Applications of focused ultrasound in the brain: from thermoablation to drug delivery

Ying Meng et al.

Summary: Focused ultrasound (FUS) is a disruptive medical technology that can precisely intervene in key circuits of brain disorders, with various forms of actions such as blood-brain barrier opening and neuromodulation. In recent years, there has been a significant expansion of FUS applications in humans, leading to increased academic and public interest in the technology.

ImmunoPET-informed sequence for focused ultrasound-targeted mCD47 blockade controls glioma

Natasha D. Sheybani et al.

Summary: Phagocytic immunotherapies, such as CD47 blockade, have shown promise in treating glioblastoma (GB). Overcoming the challenges posed by the blood brain/tumor barriers (BBB/BTB) for mCD47 delivery through focused ultrasound (FUS) is crucial. Timing of antibody injection relative to FUS BBB/BTB disruption is essential for optimal mCD47 access to gliomas. Repeat sessions of FUS can significantly constrain tumor outgrowth and extend survival in glioma-bearing mice, making mCD47 delivery with FUS a promising therapeutic strategy for GB therapy.

Sonodynamic Therapy for the Treatment of Intracranial Gliomas

Antonio D'Ammando et al.

Summary: SDT shows promise as a treatment for malignant gliomas, combining low-intensity ultrasound waves with sensitizing compounds to enhance immunogenicity, increase apoptotic rates, and reduce angiogenetic potential in tumor cells. Preclinical studies have confirmed its anti-tumoral properties and first clinical trials are set to begin recruiting patients. Further investigations are needed to explore the clinical applications of SDT for intracranial gliomas.

Role of Dynamic Susceptibility Contrast Perfusion MRI in Glioma Progression Evaluation

Guanmin Quan et al.

Summary: The study suggests that combining DSC-MRI with conventional MRI can improve the diagnostic performance for identifying true progression and pseudoprogression in glioma patients, achieving a high sensitivity and specificity. This approach may promptly distinguish these conditions without the need for long-term follow-up.

DCE-MRI in Glioma, Infiltration Zone and Healthy Brain to Assess Angiogenesis: A Biopsy Study

Vera C. Keil et al.

Summary: The study found that DCE-MRI kinetic parameters are correlated with microvascular parameters in gliomas and can be used to predict neovascularization. Differences in K-trans and v(e) values were observed in the infiltration zones of patients in different prognostic groups, and significant differences were found in vascular parameters between vital tumor tissue and infiltration zones.

Focused ultrasound mediated blood-brain barrier opening is safe and feasible in a murine pontine glioma model

Zachary K. Englander et al.

Summary: The study demonstrated that FUS is a safe and feasible technique for opening the BBB and delivering drugs to the brainstem efficiently without causing significant adverse reactions. Furthermore, FUS can significantly increase drug concentration within the tumor, providing a new direction for the treatment of pontine glioma.

Contrast-Free Detection of Focused Ultrasound-Induced Blood-Brain Barrier Opening Using Diffusion Tensor Imaging

Maria Eleni Karakatsani et al.

Summary: The study investigated the use of diffusion tensor imaging for detecting focused ultrasound-induced blood-brain barrier opening, showing an increase in fractional anisotropy and diffusion anisotropy enhancement post-BBB opening. The experiment confirmed the feasibility of the technique and suggested its potential as a contrast-free MRI modality for evaluating BBB integrity in brain-related pathologies.

Immunological Effects of Histotripsy for Cancer Therapy

Alissa Hendricks-Wenger et al.

Summary: Immunomodulating therapies have become a key approach in the treatment of cancer, addressing not only primary tumors but also circulating tumor cells and distant metastases. Focal ablation therapies, such as histotripsy, have shown promise in debulking tumors and stimulating an immune response. Studies also suggest that histotripsy may have reproducible immune effects across different tumor types.
