How different are marine microbial natural products compared to their terrestrial counterparts?

Tanja M. Voser et al.

Summary: The majority of marine microorganism natural products are closely related to those isolated from terrestrial microbes, with only a small percentage being unique. Focusing on marine specific and understudied microbial phyla increases the likelihood of discovering unique marine natural compounds.

Auyuittuqamides A-D, Cyclic Decapeptides from Sesquicillium microsporum RKAG 186 Isolated from Frobisher Bay Sediment

Alyssa L. Grunwald et al.

Summary: Four new cyclic decapeptides, auyuittuqamides A-D (1-4), were isolated from marine sediment collected from the intertidal zone of Frobisher Bay, Nunavut, Canada. The compounds' structures were elucidated using NMR spectroscopy and tandem mass spectrometry, while the absolute configurations of the amino acids were determined using Marfey's method.

Freshwater Fungi as a Source of Chemical Diversity: A Review

Tamam El-Elimat et al.

Summary: This review summarizes the chemical diversity and biological activity of 199 out of 280 known freshwater fungal metabolites published from 1992 to 2020, representing at least seven structural classes. In addition to describing the metabolites themselves, strategies for the collection, isolation, and identification of fungi from freshwater habitats are discussed to enhance chemists' knowledge of mycological principles. The review is expected to serve as a springboard for future natural products studies from the underexplored group of Ascomycota.

Leveraging Peptaibol Biosynthetic Promiscuity for Next-Generation Antiplasmodial Therapeutics

Jin Woo Lee et al.

Summary: Malaria, affecting over 200 million people annually, remains a global threat. Through systematic testing of fungal extracts from the University of Oklahoma Citizen Science Soil Collection Program, bioactive compounds with potent antiplasmodial activity and low toxicity against human cells were identified, with a new metabolite, harzianin NPDG I, showing remarkable potency and selectivity. This study highlights the potential of fungal diversity in developing novel anti-malarial compounds.

Building Natural Product Libraries Using Quantitative Clade-Based and Chemical Clustering Strategies

Victoria M. Anderson et al.

Summary: This study proposes a set of tools combining genetic barcoding and metabolomics to help researchers build natural product libraries. Through experimentation with fungi, it was found that a modest number of isolates could provide a significant portion of chemical features, with different subclades containing varying levels of chemical diversity.

Evolutionary and functional analysis of an NRPS condensation domain integrates β-lactam, D-amino acid, and dehydroamino acid synthesis

Michael J. Wheadon et al.

Summary: Nonribosomal peptide synthetases (NRPSs) are large, multidomain enzymes involved in the assembly-line-like synthesis of peptide natural products, including clinically useful antibiotics. The condensation (C) domains in NRPSs catalyze peptide bond formation during synthesis. Research has shown that in certain cases, the function of C domains can be altered, providing insights into beta-lactam synthesis and canonical peptide synthesis mechanisms.

Identical sequences found in distant genomes reveal frequent horizontal transfer across the bacterial domain

Michael Sheinman et al.

Summary: Horizontal gene transfer (HGT) is a key force in microbial evolution, with long-distance HGT being frequent. The function of sequences strongly impacts their transfer rate, varying significantly between different functional categories. This study provides a comprehensive view of HGT and its role in driving bacterial evolution.
