Synthesis and evaluation of azalamellarin N and its A-ring-modified analogues as non-covalent inhibitors of the EGFR T790M/L858R mutant

Tsutomu Fukuda et al.

Summary: Synthetic azalamellarin analogues showed higher inhibitory activity compared to lamellarin and demonstrated strong selectivity, with the analogue containing two 3-(dimethylamino)propoxy groups showing the best performance. The inhibitory activity was attributed to hydrogen bonding interaction between the lactam NH group of the B-ring and the carbonyl group of a methionine residue.

Evaluation of the binding behavior of olmutinib (HM61713) with model transport protein: Insights from spectroscopic and molecular docking studies

Song-Bo Kou et al.

Summary: S C Olmutinib is a novel third-generation tyrosine kinase inhibitor that selectively targets epidermal growth factor receptor mutations. The binding behavior of OMT with bovine serum albumin was studied, revealing hydrophobic interactions, hydrogen bonds, and van der Waals forces as key factors in the complexation process. OMT tended to bind to subdomain IB of BSA, causing changes in BSA's conformation and secondary structure as observed through fluorescence, FT-IR, and UV-Vis studies.

A Novel Thienopyrimidine Analog, TPH104, Mediates Immunogenic Cell Death in Triple-Negative Breast Cancer Cells

Diwakar Bastihalli Tukaramrao et al.

Summary: This study reports a novel compound, TPH104, that induces a unique, non-apoptotic cell death in triple-negative breast cancer (TNBC) cells by activating dendritic cells (DCs). TPH104 enhances the tumor immunogenic microenvironment to increase the efficacy of conventional chemotherapy for TNBC.

Design, Synthesis, and Antitumor Activity of Olmutinib Derivatives Containing Acrylamide Moiety

Xiaohan Hu et al.

Summary: Two series of olmutinib derivatives containing an acrylamide moiety were synthesized and evaluated for their IC50 values against five cancer cell lines, showing moderate cytotoxic activity. The most promising compound, H10, exhibited excellent activity against EGFR kinase and positive biological activity against PI3K kinase. The SAR indicated that the introduction of dimethylamine scaffolds with smaller spatial structures was more favorable for antitumor activity. Overall, compounds H7 and H10 were confirmed as promising antitumor agents.
