Contralateral Effects of Eccentric Exercise and DOMS of the Plantar Flexors: Evidence of Central Involvement

Suresh Marathamuthu et al.

Summary: This study aimed to investigate the peripheral and central factors involved in the reduction of motor performance following eccentric exercise and delayed onset muscle soreness (DOMS) in both treated and contralateral limbs. The results showed that central factors played a significant role in the reduction of force immediately following eccentric exercise and during the later stage of DOMS.

Imbalanced Subthreshold Currents Following Sepsis and Chemotherapy: A Shared Mechanism Offering a New Therapeutic Target?

Mark M. Rich et al.

Summary: Both sepsis and chemotherapy can cause neurologic dysfunction, characterized by neuropathy and encephalopathy. The underlying mechanisms are not well understood. Preclinical models reveal defects in ion channel function in both conditions, contributing to peripheral neuropathy. These defects may also extend to the central nervous system, leading to encephalopathy. Restoring the balance of subthreshold currents through pharmacological intervention could be a potential therapeutic approach for the treatment of neuropathy and encephalopathy in patients recovering from sepsis or chemotherapy.

New Perspectives in the Pathophysiology and Treatment of Pain in Patients with Dry Eye Disease

Giuseppe Giannaccare et al.

Summary: Ocular discomfort and eye pain are common symptoms in patients with dry eye disease (DED), presenting a challenge for ophthalmologists. Recent research suggests that enkephalins, endorphins, and dynorphins, which are endogenous opioid peptides, may play a role in modulating eye pain sensations. This opens up new possibilities for therapeutic approaches in managing ocular discomfort and eye pain in DED.

Pathogenic Mechanism of Dry Eye-Induced Chronic Ocular Pain and a Mechanism-Based Therapeutic Approach

Yuto Tei et al.

Summary: The pathogenic mechanism of dry eye-induced chronic pain in the anterior eye area was explored in a rat dry eye model. The study found that hypersensitivity and hyperalgesia were caused by central sensitization in the trigeminal nucleus with upregulation of the alpha(2)delta-1 subunit. Chronic treatment with pregabalin effectively alleviated the pain symptoms and normalized neural activity and astrocyte activation.

Delayed Onset Muscle Soreness (DOMS): The Repeated Bout Effect and Chemotherapy-Induced Axonopathy May Help Explain the Dying-Back Mechanism in Amyotrophic Lateral Sclerosis and Other Neurodegenerative Diseases

Balazs Sonkodi

Summary: Delayed onset muscle soreness (DOMS) and amyotrophic lateral sclerosis (ALS) are proposed to be caused by acute compression axonopathy of sensory afferents in the muscle spindle, possibly due to genetic/environmental factors, damaging contractions, and neuroinflammatory processes. The dysfunctional microcircuits in these diseases can affect memory and learning, ultimately interfering with life-sustaining central pattern generators in ALS.

Modeling of full-length Piezo1 suggests importance of the proximal N-terminus for dome structure

Jiehan Chong et al.

Summary: Using computational methods, the three-dimensional structure of full-length Piezo1 was predicted, revealing a trilobed dome that extends beyond the radius of the protein in the membrane and preferential interactions with cholesterol and PIP2 molecules. These findings suggest the importance of N-terminal regions for Piezo1 function and interactions with the lipid bilayer.

Summary: The study found that Piezo1 is abundantly expressed in the trabecular meshwork and can regulate trabecular meshwork cell contraction by modulating Ca2+ influx and release of arachidonic acid and PGE(2). This suggests that Piezo1 could act as a regulator to control intraocular pressure.

Mechanisms of Peripheral and Central Pain Sensitization: Focus on Ocular Pain

Giulia Puja et al.

Summary: Persistent ocular pain caused by corneal inflammation and nerve injury leads to significant alterations along the pain axis, with changes in both primary sensory neurons in the trigeminal nerves and secondary neurons in the spinal trigeminal nucleus contributing to peripheral and central pain sensitization. Studies using animal models have provided insights into the mechanisms involved in these maladaptive changes. New techniques such as optogenetics and genetic neuronal labelling allow for investigation of identified circuits involved in nociception at the spinal and trigeminal levels. Recent advances in the discovery of molecular and cellular mechanisms contributing to pain sensitization in the trigeminal pathways are also presented.

Does Compression Sensory Axonopathy in the Proximal Tibia Contribute to Noncontact Anterior Cruciate Ligament Injury in a Causative Way?-A New Theory for the Injury Mechanism

Balazs Sonkodi et al.

Summary: The authors propose a new mechanism for non-contact anterior cruciate ligament injury, suggesting that it is caused by acute compression microinjury of entrapped peripheral proprioceptive sensory axons in the proximal tibia. They highlight the importance of primary damage during strenuous exercises and secondary damage during deceleration. Elevated prostaglandin E2, nitric oxide, and glutamate may play critical roles in the neuro-modulation, leading to mechano-energetic failure and inflammatory events.

Summary: Post orgasmic illness syndrome is a rare condition with unknown pathomechanism and uncertain treatment, characterized by symptoms lasting 2-7 days post ejaculation. Proposed primary injury includes acute compression proprioceptive axonopathy in the muscle spindle and mitochondrial impairment. Similarities to delayed onset muscle soreness exist, but differences arise from spermidine depletion-induced physiological responses.
