Multi-combined 3D-QSAR, docking molecular and ADMET prediction of 5-azaindazole derivatives as LRRK2 tyrosine kinase inhibitors

Saida Khamouli et al.

Summary: The study performed a three-dimensional quantitative structure-activity relationships (3D-QSAR) analysis on 26 azaindazole derivatives as LRRK2 inhibitors, achieving high activity-descriptor relationship efficiency. The models were validated externally and through docking studies, confirming their reliability and providing valuable clues for designing new compounds against LRRK2. Furthermore, an analysis of the compounds' ADMET properties and drug likeness was conducted, aiding in the optimization of new anti-Parkinson drugs.

Cameroonian medicinal plants as potential candidates of SARS-CoV-2 inhibitors

Romuald Tematio Fouedjou et al.

Summary: This study conducted in vitro experiments on metabolites of certain Cameroonian medicinal plants from the Asteraceae family in order to identify potential molecules for combating COVID-19. One compound (compound 16) showed promising pharmacokinetic parameters and low toxicity, and its stability and interaction with the viral main protease protein and spike protein were confirmed through molecular dynamics simulation.

Discovery of Potent SARS-CoV-2 Inhibitors from Approved Antiviral Drugs via Docking and Virtual Screening

Samir Chtita et al.

Summary: This study used virtual screening and molecular docking methods to identify potential inhibitors from existing drugs that can effectively fight against COVID-19. A total of 129 approved drugs were analyzed, and 5 promising drugs were found to tightly bind to the main protease of the SARS-CoV-2 virus. Further in vivo testing is needed to confirm their efficacy as clinical drugs.

Samir Chtita et al.

Summary: In silico research on forty unsymmetrical aromatic disulfide derivatives as inhibitors of the SARS Coronavirus revealed the main protease of these compounds are mainly governed by five descriptors. This led to the establishment of valid and robust statistics models using the QSAR approach.

Biophysical insight into the binding mechanism of doxofylline to bovine serum albumin: An in vitro and in silico approach

Sharmin Siddiqui et al.

Summary: Insight gained into the mechanistic binding of bovine serum albumin with doxofylline revealed a spontaneous and exothermic process, along with pronounced structural changes in BSA upon binding with doxofylline. The study also explored the influence of metal ions and vitamins on the binding affinity of the BSA-doxofylline system. Additionally, both in vitro and in silico analyses supported the strong interaction between BSA and doxofylline, suggesting its potential for optimal distribution into the bloodstream for asthma treatment.

Design, synthesis and molecular modeling studies of novel mesalamine linked coumarin for treatment of inflammatory bowel disease

Biswabhusan Rath et al.

Summary: In this study, a mesalamine-coumarin derivative showed therapeutic potential for ulcerative colitis in a rat model, with higher binding affinity to key proteins and greater stability in complex formation compared to mesalamine alone. These findings suggest that the derivative may be effective in managing inflammatory bowel diseases.
