Ribonucleotide incorporation into DNA during DNA replication and its consequences

Zhi-Xiong Zhou et al.

Summary: Ribonucleotides are the most abundant non-canonical nucleotides in the genome, with significant influence on genome biology. Recent progress has been made in understanding their genomic presence, incorporation characteristics, and usefulness as biomarkers for polymerase enzymology. The discussion also covers ribonucleotide processing, genetic consequences of unrepaired ribonucleotides in DNA, and evidence supporting their transient presence in the nuclear genome.

Deleterious point mutations in T-cell acute lymphoblastic leukemia: Mechanistic insights into leukemogenesis

Urbi Roy et al.

Summary: T-cell acute lymphoblastic leukemia (T-ALL) is characterized by accumulation of genetic and epigenetic lesions leading to sustained proliferation of abnormal T cells. Driver mutations in genes disrupt genetic and epigenetic homeostasis and understanding their mechanism could pave the way for targeted therapies to improve overall survival.

Summary: DNA polymerases are crucial enzymes for DNA replication and repair. Among the X family members, DNA polymerase mu stands out for its unique ability to synthesize DNA from an unpaired primer terminus and utilize both ribonucleotides and deoxyribonucleotides as substrates during NHEJ gap-filling.

MicroRNA miR-29c regulates RAG1 expression and modulates V(D)J recombination during B cell development

Rupa Kumari et al.

Summary: The study showed that miR-29c regulates RAG1 in a B cell stage-specific manner, modulation of miR-29c levels affects V(D)J recombination efficiency in pre-B cells, and a negative correlation between miR-29c and RAG1 levels is observed in different developmental stages.
