Microbial Metabolite Inspired β-Peptide Polymers Displaying Potent and Selective Antifungal Activity

Summary: With the quick increase of serious invasive fungal infections and the limited antifungal drugs available, there is an urgent need for potent and selective antifungal agents. Inspired by microbial metabolites, the authors have designed and obtained poly(DL-diaminopropionic acid) (PDAP), a potent antifungal agent that kills fungal cells by penetrating the cytoplasm, generating reactive oxygen, and inducing fungal apoptosis. PDAP displays strong antifungal activity against Candida albicans, negligible hemolysis and cytotoxicity, and no susceptibility to antifungal resistance. In addition, PDAP effectively inhibits fungal biofilm formation and eradicates mature biofilms. In vivo studies have shown that PDAP is safe and effective in treating fungal keratitis, highlighting its potential as a promising new antifungal agent.

Superfast and Water-Insensitive Polymerization on α-Amino Acid N-Carboxyanhydrides to Prepare Polypeptides Using Tetraalkylammonium Carboxylate as the Initiator

Summary: The tetraalkylammonium carboxylate-initiated NCA polymerization allows for superfast polymerization without the need for extra catalysts and can be operated in open vessels at ambient condition. This method can easily generate block copolymers with at least 15 blocks, avoid tedious purification steps, and enable direct polymerization in aqueous environments for the preparation of polypeptides and polypeptide nanoparticles. The advantages and mild conditions of this polymerization suggest great potential for functional exploration and application of polypeptides.

Modular Design of Membrane-Active Antibiotics: From Macromolecular Antimicrobials to Small Scorpionlike Peptidomimetics
