Estimating heritability using family-pooled phenotypic and genotypic data: a simulation study applied to aquaculture

Nima Khalilisamani et al.

Summary: Estimating heritability based on individual phenotypic and genotypic measurements can be expensive and labour-intensive in commercial aquaculture breeding. This study investigated the feasibility of estimating heritability using within-family means of phenotypes and allelic frequencies. The results showed that at least 200 families of 60 progeny per family divided equally in two pools per family were required to effectively estimate the heritability of family means. Increasing the size of pools resulted in increasing the heritability of family means towards one.

Autism genes converge on asynchronous development of shared neuron classes

Bruna Paulsen et al.

Summary: This study utilized organoid models of the human cerebral cortex to identify cell-type-specific developmental abnormalities resulting from haploinsufficiency in ASD risk genes SUV420H1, ARID1B, and CHD8. The research found asynchronous development of cortical neuronal lineages and distinct molecular pathways associated with these mutations. The study revealed shared cell-type-specific neurodevelopmental abnormalities across ASD risk genes in a manner influenced by individual genomic context.

Comparing Copy Number Variations in a Danish Case Cohort of Individuals With Psychiatric Disorders

Xabier Calle Sanchez et al.

Summary: This population-based case-cohort study examined the association between recurrent genomic copy number variants (CNVs) and mental disorders, finding that CNVs were broadly associated with an increased risk of autism spectrum disorder and ADHD, while the risk estimates for schizophrenia were lower. Sex may modulate the pathogenicity of many CNVs.

Mapping genomic loci implicates genes and synaptic biology in schizophrenia

Vassily Trubetskoy et al.

Summary: In this study, a two-stage genome-wide association study was conducted to identify common variants associated with schizophrenia. The results revealed 287 distinct genomic loci and 120 genes likely to be involved in the development of schizophrenia. This research provides valuable insights into the pathophysiology of schizophrenia and offers a resource for further mechanistic studies.

Rare coding variants in ten genes confer substantial risk for schizophrenia

Tarjinder Singh et al.

Summary: Rare coding variants in certain genes, particularly those related to the function of the nervous system, are found to be significantly associated with schizophrenia. The involvement of specific receptor subunits in the glutamatergic system supports the hypothesis of dysfunction in this system as a mechanism in the development of schizophrenia. The findings suggest that both common and rare genetic risk factors are involved in the pathogenesis of schizophrenia, and more risk genes are yet to be discovered using this approach.

Summary: The Global Burden of Diseases Study in 2019 showed that mental disorders remained among the top ten leading causes of burden worldwide, with no reduction in burden since 1990. The majority of the burden in disability-adjusted life-years (DALYs) came from years lived with disability (YLDs).

Investigating rare pathogenic/likely pathogenic exonic variation in bipolar disorder

Xiaoming Jia et al.

Summary: In this study, an analysis of exonic variation in individuals with BD did not show a replicable enrichment of rare P-LP variants across the exome or in any of several groups of genes with biologic plausibility. Despite common genetic variation between BD and schizophrenia, no association was found between BD risk and rare P-LP coding variants in genes known to modulate risk for schizophrenia.

Patterns of de novo tandem repeat mutations and their role in autism

Impact of bipolar disorder on health-related quality of life and work productivity: Estimates from the national health and wellness survey in Japan

Summary: The study shows that bipolar disorder negatively affects patients' quality of life, work productivity, and indirect costs. Compared to patients with major depressive disorder, bipolar disorder patients have lower quality of life and greater work productivity loss. These findings emphasize the importance of proper screening, diagnosis, and treatment for bipolar disorder and bipolar depression.

Presymptomatic training mitigates functional deficits in a mouse model of Rett syndrome

Nathan P. Achilly et al.

Summary: Research using a mouse model of Rett syndrome shows that intensive training before symptom onset can significantly improve specific motor and memory tasks, delaying the onset of symptoms. The study indicates that task-specific neurons activated during training develop more dendritic arbors and have better neurophysiological responses, enhancing their functionality and delaying symptom onset.

Protein-coding repeat polymorphisms strongly shape diverse human phenotypes

Ronen E. Mukamel et al.

Summary: This study utilized whole-exome sequencing data to estimate VNTR lengths and impute VNTR alleles into single-nucleotide polymorphism haplotypes, identifying strong associations of common variants with human phenotypes like height, hair morphology, and biomarkers of health. Accounting for large-effect VNTRs enabled further fine-mapping of associations to many more protein-coding mutations in the same genes. These findings point to hidden effects of highly polymorphic common structural variants that have not been previously analyzed at the molecular level.
