MafA, NeuroD1 and HNF1β synergistically activate the Slc2a2 (Glut2) gene in β-cells

Summary: The study identified an evolutionarily conserved region located approximately 40 kbp downstream of the transcription start site of the Slc2a2 gene as an active enhancer in the MIN6 beta-cell line. Three beta-cell-enriched transcription factors, MafA, NeuroD1, and HNF1 beta, were found to synergistically activate transcription through this enhancer and the proximal promoter region of the gene. Co-immunoprecipitation experiments suggested that these transcription factors interact with each other and are critical for Slc2a2 gene expression.

Saturated fatty acids entrap PDX1 in stress granules and impede islet beta cell function

Summary: This study revealed that saturated fatty acids disrupt nucleocytoplasmic transport and induce stress granule (SG) formation in pancreatic beta cells. The formation of SGs is mediated by the phosphoinositide 3-kinase (PI3K)/eukaryotic translation initiation factor 2 alpha (EIF2 alpha) pathway, and may be linked to beta cell dysfunction. Disruption of SG assembly may be a potential therapeutic strategy for treating obesity and type 2 diabetes.

Summary: Our study demonstrated that abnormal NKX6.1 expression in beta cells, with cells showing no nuclear NKX6.1 expression but insulin expression and cells showing cytoplasmic NKX6.1 expression but no insulin expression, significantly increased in type 2 diabetic subjects and were correlated with glycated hemoglobin levels, indicating elevated beta cell dedifferentiation in T2DM. The initiation of beta cell dedifferentiation might mainly occur after T2DM diagnosis, rather than in subjects with higher risks for developing T2DM.
