Rate of recipient-derived alveolar macrophage development and major histocompatibility complex cross-decoration after lung transplantation in humans

Mark E. Snyder et al.

Summary: This study revealed a biphasic rate in alveolar macrophages turnover post lung transplantation, with recipient-derived lung macrophages repopulating the lung mainly in large clusters. The recipient alveolar macrophages were shown to take up and present donor peptide-MHC complexes, but were unable to independently induce an in vitro alloreactive response by circulating recipient T cells.

Type-1 immunity and endogenous immune regulators predominate in the airway transcriptome during chronic lung allograft dysfunction

Carlo J. Iasella et al.

Summary: Chronic lung allograft dysfunction (CLAD) is a major complication limiting long-term survival among lung transplant recipients, and the airway transcriptome may reflect key immunologic changes in the disease.

Human Lung-Resident Macrophages Colocalize with and Provide Costimulation to PD1hi Tissue-Resident Memory T Cells

Mark E. Snyder et al.

Summary: EVLP is used to study the spatial and functional relationship between human lung tissue-resident memory T cells and lung-resident macrophages. Human lung T-RM and M-LR colocalize preferentially around the airways, and CD8(+) T(RM) show two functionally distinct populations based on PD1 and ZNF683 protein expression. M-LR provide costimulation signaling to T-RM, enhancing their effector function.

Resident memory T cells form during persistent antigen exposure leading to allograft rejection

Khodor Abou-Daya et al.

Summary: Research shows that in organ transplants, recipient effector T cells differentiate into functional graft T-RM that maintain rejection locally. The majority of T-RM lack exhaustion markers, serving as a platform for the discovery of therapeutic targets.

CRIP1 cooperates with BRCA2 to drive the nuclear enrichment of RAD51 and to facilitate homologous repair upon DNA damage induced by chemotherapy

Huiying Sun et al.

Summary: The study reveals that CRIP1 downregulation in gastric cancer cells results in HR repair deficiency and increased sensitivity to various chemotherapeutic drugs. CRIP1 promotes HR repair through mechanisms such as stabilizing BRCA2 and promoting nuclear enrichment of RAD51. High CRIP1 expression in patient samples correlates with poor prognosis in response to genotoxic treatments in various solid tumors.
