Engineering Natural Competence into the Fast-Growing Cyanobacterium Synechococcus elongatus Strain UTEX 2973

Kristen E. Wendt et al.

Summary: Natural transformation, a process by which bacteria uptake and incorporate extracellular DNA, plays a significant role in bacterial evolution. Cyanobacteria, serving as important model systems for studying photosynthesis, have not been extensively characterized for natural transformation. By introducing polymorphic alleles, a naturally transformable and fast-growing cyanobacterial strain was created.

Salt-Tolerant Synechococcus elongatus UTEX 2973 Obtained via Engineering of Heterologous Synthesis of Compatible Solute Glucosylglycerol

Jinyu Cui et al.

Summary: The salt tolerance of the cyanobacterium Syn2973 was successfully improved by introducing the glucosylglycerol (GG) biosynthetic pathway, overexpressing glycerol-3-phosphate dehydrogenase, and downregulating the gene rfbA. The growth of the engineered strain M-2522-GgpPS-drfbA was improved by 62% compared to the control strain M-pSI-pSII under 0.5M NaCl treatment at 60 hours. Comparative metabolomic analysis showed that more carbon flux was redirected from ADP-GLC to GG synthesis in the engineered strain, providing important engineering strategies for improving salt tolerance and GG production in Syn2973.

The Dual Role of the Glycolipid Envelope in Different Cell Types of the Multicellular Cyanobacterium Anabaena variabilis ATCC 29413

Ritu Garg et al.

Summary: Anabaena variabilis is a filamentous cyanobacterium capable of differentiating into specialized cells, heterocysts and akinetes, to survive under different stress conditions. Both cell types are characterized by the presence of a thick multilayered envelope, which includes a glycolipid layer. Studies have shown that heterocysts and akinetes use a glycolipid of identical chemical composition in their special envelopes, highlighting the importance of the glycolipid layer for their functions.

Exploiting Polyploidy for Markerless and Plasmid-Free Genome Engineering in Cyanobacteria

Christopher M. Jones et al.

Summary: The study introduces a novel plasmid-free genome engineering approach in cyanobacteria, utilizing the polyploidy of their chromosomes to create stable and unmarked mutants efficiently. By integrating DNA modifying enzyme genes into essential genes on the chromosome and maintaining antibiotic selection, genetic editing can be achieved and the modifying enzyme gene can be promptly removed post-editing. This method demonstrates reusability and flexibility in constructing mutants without the need for additional shuttle vectors or counterselection systems.

Summary: Using only three ONT flow cells, thousands of organisms, including bacteriophages, were identified, with a large part at species level. Genomes could be assembled from environmental samples with Flye, resulting in >1 Mbp contigs in several cases and a near complete genome for a Thioglobus singularis species. k-mer analysis revealed that a significant portion of the data represented species not yet deposited in the database. This approach is suitable for scalable genomic investigations and provides a new platform for education in biodiversity.
