Conditional and tissue-specific approaches to dissect essential mechanisms in plant development

Marie L. Pfeiffer et al.

Summary: Reverse genetics approaches are widely used for studying gene function, but severe mutations can limit functional studies in specific environments. The CRISPR technology has revolutionized targeted genome modification in plants, enabling inducible and tissue-specific manipulation of gene function at the DNA and RNA levels. Additionally, novel approaches for directly manipulating target proteins have been introduced in plant systems.

Identification and Transcriptional Analysis of Zinc Finger-Homeodomain (ZF-HD) Family Genes in Cucumber

Wei Lai et al.

Summary: This study conducted a comprehensive analysis of the ZF-HD gene family in cucumber for the first time, revealing their crucial roles in regulating plant growth, development, and stress responses. The results lay the foundation for further functional analysis of ZF-HD genes and exploring their potential application in improving stress tolerance in cucumber.

Genome-wide identification of ZF-HD gene family in Triticum aestivum: Molecular evolution mechanism and function analysis

Hongli Niu et al.

Summary: The study conducted the whole genome identification and expression analysis of the ZF-HD gene family in wheat for the first time, identifying 37 TaZF-HD genes which were divided into TaMIF and TaZHD subfamilies based on conserved domains. The genes were distributed across multiple chromosomes and showed powerful purification selection pressure during wheat evolution. Additionally, qRT-PCR analysis revealed significant expression patterns of TaZF-HD genes under various stress conditions, and subcellular localization experiments indicated distinct locations for specific genes within the cell.

Highly efficient multiplex editing: one-shot generation of 8x Nicotiana benthamiana and 12x Arabidopsis mutants

Johannes Stuttmann et al.

Summary: Genome editing using RNA-guided nucleases like SpCas9 has been successful in various plant species. A toolkit based on an optimized zCas9i gene allows targeting multiple independent loci simultaneously. Efficient multiplexing was achieved in Nicotiana benthamiana and Arabidopsis thaliana, with the latter showing lower efficiency due to Cas9 availability limitations. This study opens new possibilities for generating complex genotypes through multiplexing.

Systematic Investigations of the ZF-HD Gene Family in Tobacco Reveal Their Multiple Roles in Abiotic Stresses

Jinhao Sun et al.

Summary: ZF-HD transcription factors are important for plant growth and stress responses. The study identified 32 ZF-HD genes in tobacco, with NtZF-HD21 showing significant response to drought stress. Subcellular localization analysis and gene silencing experiments demonstrated that NtZF-HD21 is a nuclear protein with potential function in drought resistance.
