Improving the catalytic efficiency and dimeric stability of Cu,Zn superoxide dismutase by combining structure-guided consensus approach with site-directed mutagenesis

Sachin Kumar et al.

Summary: This study utilized a structure-guided consensus approach to improve the enzymatic properties of Cu,Zn SOD from the high-altitude plant Potentilla atrosanguinea. The T97D substitution in SOD led to improved dimer stability and catalytic efficiency compared to the wild-type (WT), as validated by computational and experimental tools. This mutation not only enhanced enzyme thermostability but also laid the groundwork for designing SODs with improved kinetics by exploring interface residues as potential targets.

Evaluation of acridinedione analogs as potential SARS-CoV-2 main protease inhibitors and their comparison with repurposed anti-viral drugs

Vijay Kumar Bhardwaj et al.

Summary: The study identified DSPD-2, DSPD-6, and DSPD-5 as potential inhibitors of SARS-CoV-2 by efficiently binding to the S1 subsite of Mpro. Targeting molecules to bind effectively to the S1 subsite could potentially improve the binding of molecules to SARS-CoV-2 Mpro, suggesting a new approach for developing antiviral drugs.

Explicit-solvent molecular dynamics simulations revealed conformational regain and aggregation inhibition of I113T SOD1 by Himalayan bioactive molecules

Sachin Kumar et al.

Summary: The study suggests that Himalayan bioactive molecules can effectively revert the structural changes caused by I113T, preventing aggregation and potentially serving as therapeutic molecules for ALS. Further validation through in vitro and in vivo studies is needed to confirm these findings.

Site-directed mutagenesis (P61G) of copper, zinc superoxide dismutase enhances its kinetic properties and tolerance to inactivation by H2O2

Sachin Kumar et al.

Summary: The engineered Cu, Zn SOD (WT) with a substitution of a conserved residue at position 61 (P61G) showed higher catalytic efficiency and thermostability compared to the wild-type protein. The P61G mutation also resulted in enhanced tolerance to hydrogen peroxide inactivation, making it a potential candidate for applications in the food and pharmaceutical industries.

Concurrent circulation of dengue serotype 1, 2 and 3 among acute febrile patients in Cameroon

Huguette Simo Tchetgna et al.

Summary: Between July and December 2020, acute febrile patients in Douala, Cameroon were screened for dengue infections, with 12.8% testing positive. DENV-3 was the most common serotype, followed by DENV-2 and DENV-1. Co-infections with malaria were also observed in 56% of dengue cases. Further epidemiological studies are needed in major cities of the region to address dengue as a serious public health threat.

Taming the ringmaster of the genome (PCNA): Phytomolecules for anticancer therapy against a potential non-oncogenic target

Vijay Kumar Bhardwaj et al.

Summary: PCNA is a crucial nuclear protein involved in DNA replication and repair, often referred to as the ringmaster of the genome. Computational screening identified Hypericin and Pseudohypericin from Hypericum perforatum as potential inhibitors of PCNA with stronger potency than standard inhibitors. Further experimental studies are needed to validate these findings.
