The histone demethylase PHF8 regulates TGFβ signaling and promotes melanoma metastasis

Rana S. Moubarak et al.

Summary: In this study, the researchers identified PHF8 as a previously unidentified prometastatic gene in melanoma. They found that PHF8 promotes cell invasion and is involved in the TGF beta signaling pathway, providing new insights into the epigenetic regulation of metastasis in cancer.

Histone demethylase PHF8 drives neuroendocrine prostate cancer progression by epigenetically upregulating FOXA2

Qiuli Liu et al.

Summary: The study demonstrates that PHF8 is essential for the development of NEPC by transcriptionally upregulating the FOXA2 gene, which regulates the expression of genes involved in NEPC development. Both PHF8 and FOXA2 could potentially serve as biomarkers for NEPC and targeting them may be a therapeutic strategy for NEPC treatment.

WLS-Wnt signaling promotes neuroendocrine prostate cancer

Tyler Bland et al.

Summary: Experimental studies revealed that the Wnt secretion mediator Wnt less (WLS) plays a major role in NEPC pathogenesis and aggressive tumor growth, being suppressed by AR and activating the ROR2/PKC delta/ERK signaling pathway. Clinical sample analysis showed high expression of WLS in CRPC and NEPC tumors, and treatment with WLS inhibitor LGK974 can restrict NE prostate tumor growth.

Lgr4 Regulates Oviductal Epithelial Secretion Through the WNT Signaling Pathway

Xue Tan et al.

Summary: The WNT signaling pathway plays a crucial role in the development of fallopian tubes in mice, with LGR4 regulating epithelial secretion and affecting tube size and cell number. This regulation is mediated by modulation of the WNT pathway and changes in gene expression profiles of epithelial cells. Additionally, activation of the WNT pathway triggers the expression of steroidogenic genes through the transcription factor NR5A2, facilitating epithelial secretory cell maturation and steroidogenesis for fallopian tube development and function.
