A versatile platform for generating engineered extracellular vesicles with defined therapeutic properties

Kevin Dooley et al.

Summary: Extracellular vesicles (EVs) play a crucial role in intercellular communication by facilitating the transfer of macromolecules between cells. This study identified PTGFRN and BASP1 as scaffold proteins that enable high-density loading of a wide range of molecules into EVs, demonstrating potent therapeutic potential in animal models. This engineering platform provides a simple approach to functionalize EVs with diverse macromolecules, representing a significant step toward unlocking the therapeutic potential of EVs.

Jumping Ahead with Sleeping Beauty: Mechanistic Insights into Cut-and-Paste Transposition

Matthias T. Ochmann et al.

Summary: The Sleeping Beauty transposon system has been widely used as a genetic engineering tool, with extensive studies elucidating the molecular mechanisms and actions during SB transposition process. Structural data on the SB transposase provide a direct insight into the enzyme's functionality, leading to the development of novel SB variants for advanced genetic engineering possibilities. However, many aspects of transposition process remain poorly understood, necessitating further research to explore the structure-function relationships of SB transposition for the development of new vector systems.

Exosome-Mediated Delivery of the Neuroprotective Peptide PACAP38 Promotes Retinal Ganglion Cell Survival and Axon Regeneration in Rats With Traumatic Optic Neuropathy

Tian Wang et al.

Summary: Traumatic optic neuropathy (TON) is characterized by optic nerve damage resulting in vision loss, with limited treatment options. Pituitary adenylate cyclase-activating polypeptide 38 (PACAP38) has shown promise as a therapeutic agent. Using rat retinal ganglion cell (RGC) exosomes as delivery vesicles for PACAP38 (EXOPACAP38) demonstrated improved uptake efficiency and potential for promoting RGC survival, nerve fiber layer thickness, axon regeneration, and optic nerve function in a rat TON model. EXOPACAP38 may offer therapeutic implications for TON patients.

Choice of selectable marker affects recombinant protein expression in cells and exosomes

Chenxu Guo et al.

Summary: The choice of selectable marker gene significantly impacts the level of recombinant protein expression and cell-to-cell variability in transgenic mammalian cells. Different combinations of selectable markers and antibiotics establish thresholds below which no cell can survive, and these thresholds vary significantly between different selectable markers.

Mammalian retrovirus-like protein PEG10 packages its own mRNA and can be pseudotyped for mRNA delivery

Michael Segel et al.

Summary: Eukaryotic genomes contain domesticated genes from integrating viruses and mobile genetic elements, including mammalian Gag homologs and PEG10, which can be used for RNA delivery. The development of selective endogenous encapsidation for cellular delivery (SEND) using PEG10 demonstrates its potential as an efficient therapeutic delivery modality.

Exosome-mediated delivery of an anti-angiogenic peptide inhibits pathological retinal angiogenesis

Xue Dong et al.

Summary: The study utilized exosomes (EXOs) as carriers for delivering the anti-angiogenic peptide KV11 via retro-orbital injection, showing enhanced suppression of neovascularization and vascular leakage in the retina. This system has the potential to treat proliferative retinopathy in a less invasive manner compared to intravitreal injection.
