ClpXP-mediated Degradation of the TAC Antitoxin is Neutralized by the SecB-like Chaperone in Mycobacterium tuberculosis

Pauline Texier et al.

Summary: Bacterial toxin-antitoxin systems, including TAC modules with SecB-like chaperones, are widespread in bacterial genomes. Deletion of endogenous AAA(+) proteases bypasses the need for a dedicated chaperone, and the ClpXP1 P2 complex is identified as the main protease involved in TAC antitoxin degradation.

Global Inventory of ClpP- and ClpX-Regulated Proteins in Staphylococcus aureus

Volker C. Kirsch et al.

Summary: This study investigated the functional roles of ClpP and ClpX in controlling pathogenesis in Staphylococcus aureus, utilizing genomic deletion mutants and an inactive ClpP active-site mutant strain. The results revealed downregulation of virulence, purine/pyrimidine biosynthesis, iron uptake, and stress response in mutant strains compared to the wildtype, with implications for the development of novel virulence inhibitor classes.

Contribution of the Clp Protease to Bacterial Survival and Mitochondrial Homoeostasis

Astrid Illigmann et al.

Summary: The Clp protease system plays a crucial role in bacterial survival by ensuring fast adaptation to environmental changes. It is responsible for protein homeostasis, quality control, and targeted proteolysis, and is essential for regulating various cellular processes in both prokaryotes and eukaryotes. This system not only helps bacteria cope with stress conditions and regulate developmental programs, but also plays a vital role in pathogenic species by controlling virulence factor expression and host colonization.
