Whole-cell catalytic synthesis of 2-O-α-glucopyranosyl-L-ascorbic acid by sucrose phosphorylase from Bifidobacterium breve via a batch-feeding strategy

Summary: This study identified Bifidobacterium breve's SPase as a new AA glycosylating tool with optimal conditions. Using sucrose as the glycosyl donor, it was found that fructose seriously inhibited transglycosylation activity, but a new sucrose batch-feeding strategy was successfully developed to achieve high production of AA-2G.

This study proposes using the concept of stratification to improve reliability by considering a set of states and system transitions. Developmental and neurobehavioral assessment of low-dose N-nitrosodimethylamine (NDMA) using zebrafish embryo bioassay

Parallel molecular mechanisms for enzyme temperature adaptation

Summary: This study reveals the molecular and evolutionary mechanisms of enzyme temperature adaptation, identifying that temperature adaptation in KSI primarily comes from a single residue change with limited, local epistasis. Residues associated with organismal growth temperature are also found across diverse bacterial enzyme families, suggesting widespread parallel adaptation to temperature. The study assesses residue properties, molecular interactions, and interaction networks that appear to underly temperature adaptation.

Engineering of a Thermostable Biocatalyst for the Synthesis of 2-O-Glucosylglycerol

In this study, a thermostable phosphorylase was engineered to improve its efficiency and stability for producing 2-O-Glucosylglycerol, resulting in a variant with increased catalytic efficiency, improved regioselectivity, and retained activity at elevated temperatures.

Summary: An alpha-glucosidase from Oryza sativa was heterologously produced in Pichia pastoris and used for the efficient biosynthesis of AA-2G. The purified enzyme displayed high specific activity at 37 degrees C and pH 4.0, and successfully synthesized AA-2G under specific conditions. These studies provide a foundation for the industrial application of alpha-glucosidase.
