Skin pharmacokinetics of diclofenac and co-delivered functional excipients

M. Alice Maciel Tabosa et al.

Summary: The role of excipients in the partitioning and diffusion of drugs in the skin is crucial in the development of dermatological drug products. This study correlated the skin pharmacokinetics of diclofenac and the co-uptake of two key excipients, propylene glycol and butylene glycol, in human subjects. The results highlight the importance of considering the disposition of excipients in the optimization of drug delivery from topical formulations.

In Vitro-In Vivo Correlation in Dermal Delivery: The Role of Excipients

Avnish Patel et al.

Summary: The composition of topical and transdermal formulations plays a crucial role in determining the drug delivery rate and extent through the skin. This study investigated the effects of commonly used excipients on the percutaneous absorption of ibuprofen, finding PG, DPG, and TPG to be more effective than PEG 300 in drug delivery. Quantitative CRS showed a positive correlation with in vitro diffusion studies, indicating its potential as a powerful tool for evaluating drug and vehicle delivery from dermal formulations.

Relating transdermal delivery plasma pharmacokinetics with in vitro permeation test (IVPT) findings using diffusion and compartment-in-series models

Xin Liu et al.

Summary: This study successfully related in vivo pharmacokinetic profiles to IVPT findings for topical products, showing that IVPT data can be used to accurately predict in vivo behavior of drugs. The results suggest that mechanistic and physiologically based pharmacokinetic models can be effectively utilized for predicting in vivo behavior from IVPT data for topical products.

Physiologically-based pharmacokinetic modeling to support bioequivalence and approval of generic products: A case for diclofenac sodium topical gel, 1%

Eleftheria Tsakalozou et al.

Summary: Using quantitative methods and modeling, such as PBPK modeling, can support alternative approaches to establish bioequivalence for dermatological drug products, reducing or eliminating the need for human testing. Additionally, modeling can effectively assess the similarity and bioequivalence between generic and reference products.

Predicting topical drug clearance from the skin

Maria Alice Maciel Tabosa et al.

Summary: The study shows that drug clearance from the difficult-to-access compartment in the skin can be inferred from systemic pharmacokinetic data, and is closely related to key physical chemical properties of the drug. A multiple regression analysis was used to determine this relationship and demonstrated that the model predictions correlated well with in vitro skin experiments.
