Emodin reduces tumor burden by diminishing M2-like macrophages in colorectal cancer

Alexander T. Sougiannis et al.

Summary: Our study confirms the effectiveness of emodin as a primary therapy against genetic and chemically induced sporadic colorectal cancer. We found that emodin reduces the number of protumorigenic M2-like macrophages in the tumor microenvironment. Furthermore, we provide evidence that emodin may be acting to antagonize the P2X7 receptor within the bone tissue, leading to decreased activation of proinflammatory cells and potentially affecting the recruitment of cells to the tumor microenvironment.

The role of P2X7 receptor in infection and metabolism: Based on inflammation and immunity

Xiaoyu Cai et al.

Summary: The P2X7 receptor, a ligand-gated receptor in the P2 receptor family, plays a crucial role in regulating physiological functions and disease development in the human body. It is mainly expressed in immune cells and can be activated by nucleotides and non-nucleotide substances, impacting inflammation and immunity. Targeting the P2X7 receptor may be an effective approach for treating inflammatory and immune diseases, while its role in microbial infection and metabolism holds promise for future research.

Role of the global gut microbial community in the development of colitis-associated cancer in a murine model

Jae Gon Lee et al.

Summary: The gut microbiota plays an important role in the development of colitis-associated cancer (CAC). In a murine model, altering the microbial community with various antibiotics can suppress tumorigenesis and inflammation. Combination antibiotic therapy was most effective at suppressing tumorigenesis and inflammation, while individual antibiotic treatments had different effects.

Summary: The cumulative impact of chronic inflammation in patients with inflammatory bowel diseases predisposes to the development of IBD-CRC. Inflammation can induce mutagenesis, and the molecular pathogenesis of IBD-CRC is well described, while the immunopathogenesis is less understood. The impact of novel immunosuppressive therapies on achieving deep remission in IBD-CRC remains mostly unknown.

Summary: This study identified the role of kynurenine (Kyn) and indoleamine 2,3-dioxygenase (IDO) in regulating tumor-infiltrating T cells and adaptive immunity in colorectal cancer (CRC). Inhibition of IDO diminished Kyn/AhR-mediated regulatory T cell (Treg) differentiation, providing a potential strategy for the prevention and treatment of inflammation-related colon cancer.
