The role of mitochondria in cocaine addiction

Claire Thornton et al.

Summary: The incidence of cocaine abuse is increasing, particularly in the U.K., with profound effects on brain metabolism and mitochondrial function. Cocaine exposure alters metabolic gene expression and protein networks in key brain regions, potentially impacting brain development and the pathophysiology of addiction. Therapies aimed at maintaining mitochondrial functional integrity may hold promise in mitigating cocaine addiction and pathology.

Altered cardiac mitochondrial dynamics and biogenesis in rat after short-term cocaine administration

Summary: The study demonstrates that cocaine induces alterations in cardiac mitochondrial dynamics, biogenesis, and UPRmt before any structural changes are observable in the myocardium, potentially reflecting a compensatory mechanism of cardiac mitochondria against the cardiotoxic effects of cocaine.

Lymphoblastoid Cell Lines as Models to Study Mitochondrial Function in Neurological Disorders

Sarah Jane Annesley et al.

Summary: The study of mitochondrial function in neurological disorders is challenging due to the inaccessibility of brain tissue, but the use of lymphoblastoid cell lines (LCLs) is an effective approach. These cell models can help understand mitochondrial defects, signaling pathways, and biomarkers in neurological disorders, and are valuable for the development of diagnostic tests and identification of drug targets.

Cocaine-induced neuron subtype mitochondrial dynamics through Egr3 transcriptional regulation

Shannon L. Cole et al.

Summary: Cocaine exposure affects mitochondrial dynamics and morphology by regulating transcription of mitochondria-related nuclear gene transcripts through Egr3. Decreased Egr3 expression in D1-MSNs can attenuate cocaine-induced effects on mitochondrial transcriptional coactivators and morphological adaptations, suggesting a role for Egr3 in neuronal function and plasticity with cocaine exposure.
