Peritoneal restoration by repurposing vitamin D inhibits ovarian cancer dissemination via blockade of the TGF-β1/thrombospondin-1 axis

Kazuhisa Kitami et al.

Summary: The study found that vitamin D inhibited the transformation of ovarian cancer cells into mesothelial cells and restored the normal state of mesothelial cells, thereby suppressing ovarian cancer dissemination. Vitamin D suppressed thrombospondin-1 expression by competing with Smad3, which played a significant role in preventing ovarian cancer dissemination.

Indirect co-culture of lung carcinoma cells with hyperthermia-treated mesenchymal stem cells influences tumor spheroid growth in a collagen-based 3-dimensional microfluidic model

Nandini Dhiman et al.

Summary: The study utilized a 3D tumor-on-chip device to co-culture human amniotic membrane-derived MSCs with lung carcinoma cells. Results showed that AMMSC-CM led to tumor spheroid proliferation, while heat-AMMSC-CM reduced spheroid diameter and cell proliferation. The medium containing the P1 peptide exhibited the least cytotoxicity to tumor spheroids in co-culture.

An Embryonic Diapause-like Adaptation with Suppressed Myc Activity Enables Tumor Treatment Persistence

Eugen Dhimolea et al.

Summary: Our study reveals that treatment-persistent tumor cells in cancer patients adopt a reversible transcriptional program similar to embryonic diapause, which allows them to resist chemotherapy-induced cytotoxicity. Inhibiting Myc activity or interfering with the diapause-like adaptation could be potential therapeutic strategies against chemotherapy-persistent tumor cells.

Constitutive activation of integrin αvβ3 contributes to anoikis resistance of ovarian cancer cells

Romana Dolinschek et al.

Summary: The study investigated the mechanisms of survival in ascitic ovarian cancer cells and found that a specific variant of the αvß3 integrin can affect tumor cell resistance to anoikis and cisplatin. This variant promotes activation of survival signaling pathways, inhibiting apoptosis and impacting peritoneal metastasis.

Exploring the clinical value of tumor microenvironment in platinum-resistant ovarian cancer

Alia Ghoneum et al.

Summary: Platinum resistance in high grade serous ovarian cancer is alarmingly increasing, with low five-year survival rates. The interactions between cancer cells and components of the tumor microenvironment play a crucial role in platinum resistance, along with immune-cell players. Clinical relevance of resistant markers and immunotherapies are important in combating platinum resistance in ovarian cancer.

Summary: It has been found that substances released by activated platelets may protect tumor cells from chemotherapy by promoting the expansion of ovarian cancer stem cells, thus reducing the effectiveness of chemotherapy drugs and increasing the risk of tumor relapse.

Pro-tumoral behavior of omental adipocyte-derived fibroblasts in tumor microenvironment at the metastatic site of ovarian cancer

Summary: The study demonstrated that OvCa cells induce dedifferentiation of peritoneal adipocytes, resulting in the generation of O-ADFs with pro-tumoral characteristics. Inhibition of Wnt signaling pathway blocked the dedifferentiation effect of malignant ascites on adipocytes.

Evolution of core archetypal phenotypes in progressive high grade serous ovarian cancer

Aritro Nath et al.

Summary: The study reveals the core archetypes in progressive HGSOC and demonstrates consistent enrichment of subclones with the metabolism and proliferation archetype as resistance is acquired to multiple lines of therapy. The shifts in metabolism and proliferation are associated with treatment response and clonal heterogeneity in HGSOC.

A large-scale multi-institutional study evaluating prognostic aspects of positive ascites cytology and effects of therapeutic interventions in epithelial ovarian cancer

Summary: Positive ascites cytology is a negative prognostic factor for patients with epithelial ovarian cancer, with chemotherapy potentially mitigating its impact on patient outcomes. However, complete staging surgery did not show an improvement in prognosis for patients with positive ascites cytology.

Malignant Ascites in Ovarian Cancer: Cellular, Acellular, and Biophysical Determinants of Molecular Characteristics and Therapy Response

Brittany P. Rickard et al.

Summary: The abnormal accumulation of fluid in the abdomen, known as ascites, often indicates underlying tissue dysfunction or disease, such as cancer. Among ovarian cancer patients, the presence of ascites is associated with the poorest outcomes. Despite limited understanding of the role of malignant ascites in promoting metastasis and therapy resistance, treatment options are being explored to improve patient prognosis.

Cancer-associated mesothelial cells promote ovarian cancer chemoresistance through paracrine

Jin Qian et al.

Summary: Ovarian cancer-associated mesothelial cells play a critical role in promoting chemoresistance and stemness in ovarian cancer. The secreted factor osteopontin drives mesothelial-mediated chemoresistance and stemness, and therapeutic targeting of osteopontin may enhance platinum sensitivity in ovarian cancer.
