Patients with systemic sclerosis show phenotypic and functional defects in neutrophils

Daniela Impellizzieri et al.

Summary: Neutrophils in SSc patients show functional defects in cell migration, NET formation, and phagocytosis of bacteria, with lower expression of CD16 and CD62L, higher levels of pSTAT3 and pSTAT6, lack of CXCR1 and CXCR2 receptors, and deficiency in MPO levels.

L-Selectin expression is associated with inflammatory microenvironment and favourable prognosis in breast cancer

Sarita Kumari et al.

Summary: SELL expression is significantly upregulated in breast tumors, regulated by DNA methylation, and associated with an inflammatory microenvironment. Single cell sequencing data shows similar levels of SELL expression in B-cells and T-cells in breast cancer. High SELL expression is associated with better survival outcomes in basal, Her2+and luminal B subtypes of breast cancer.

Anti-PM/Scl Antibody-positive Systemic Sclerosis Complicated by Multiple Organ Involvement

Tatsuya Shimizu et al.

Summary: A 40-year-old Japanese woman developed multiple complications, including malignant-phase hypertension, renal disease, and pulmonary arterial hypertension. Further analysis confirmed a diagnosis of anti-PM/Scl antibody-positive systemic sclerosis. The presence of anti-PM/Scl antibodies throughout the clinical course suggests a connection to multiple organ diseases, emphasizing the importance of careful observation of antinuclear antibody patterns and dermatological findings for diagnostic clarity.

Regulation of Monocyte Adhesion and Type I Interferon Signaling by CD52 in Patients With Systemic Sclerosis

Michal Rudnik et al.

Summary: The study found an increase in adhesion and type I IFN-related genes in monocytes from SSc patients, with up-regulated expression of CD11b/CD18, reduced expression of CD52, and enhanced adhesion properties. Changes in CD52 expression were consistent with SSc subtypes, immunosuppressive treatments, autoantibody profiles, and monocyte adhesion in patients. Overexpression of CD52 decreased CD18 and monocyte adhesion, while knockdown of CD52 increased adhesion. Monocytic CD52 expression was regulated by IL-4/IL-13 and down-regulated by lipopolysaccharide and IFNs alpha, beta, and gamma. Targeting the IFN-HDAC-CD52 axis in monocytes may be a new therapeutic option for early SSc patients.
