Sperm IZUMO1 Is Required for Binding Preceding Fusion With Oolemma in Mice and Rats

Takafumi Matsumura et al.

Summary: Fertilization is a complex process that involves multiple steps. The IZUMO1 protein has been shown to be essential for sperm-egg fusion in mice, but its role in other species is largely unknown. In this study, Izumo1 knockout rats were generated using CRISPR/Cas9 technology and found to be infertile. Unlike in mice, the knockout rat spermatozoa were unable to bind to the egg plasma membrane. Further investigation revealed a decrease in the number of acrosome-reacted sperm bound to the egg plasma membrane in the knockout mice. However, the binding of acrosome-reacted sperm to the egg plasma membrane was not affected in mice lacking other recently discovered fusion-indispensable genes. These findings suggest that IZUMO1 is required for sperm-oolemma binding prior to fusion, at least in rat.

Arylsulfatase A Remodeling during Human Sperm In Vitro Capacitation Using Field Emission Scanning Electron Microscopy (FE-SEM)

Maria Jose Gomez-Torres et al.

Summary: Capacitation induces changes in sperm surface ARSA density and redistribution, with a progressive clustering in the peri-acrosomal region over time, indicating a time-dependent capacitation-mediated translocation of ARSA residues on sperm surfaces. These findings offer new insights into the molecular events preceding sperm-oocyte interactions.

Bioinformatic characterization of the Transmembrane protein95 gene (TMEM95) in Murrah buffalo (Bubalus bribalis)

Sampathirao Shireesha et al.

Summary: Research on TMEM95 gene in cattle and buffalo revealed a 2 bp deletion in Murrah buffalo causing missense mutations in functional isoforms and absence of HIT ZF motif. This suggests that the subfertility gene TMEA195 in cattle and buffalo may have evolved with different functions but still play a role in male fertility.

Immunolocalization of Fertilin beta, IZUMO1, and P34H in Ram Spermatozoa

Gul Ipek Gundogan et al.

Summary: This study aimed to investigate changes in the localization of three proteins in ram spermatozoa under various conditions, suggesting that fertilin beta and P34H could potentially serve as markers for fertility or freezability in sperm. The study used different methods such as immunofluorescence labeling and Western blot analysis to compare protein expression in fresh and frozen-thawed sperm samples, highlighting the impact of freezing on sperm membrane integrity and acrosome structure.

Evolutionarily conserved sperm factors, DCST1 and DCST2, are required for gamete fusion

Naokazu Inoue et al.

Summary: The study demonstrates the essential role of evolutionarily conserved factors DCST1 and DCST2 in sperm-egg fusion in mice, and how the protein stability of SPACA6 is regulated differently by DCST1/2 and IZUMO1. This suggests that spermatozoa ensure proper fertilization in mammals by integrating various molecular pathways, including an evolutionarily conserved system developed over nearly one billion years of evolution.
