Endocannabinoids Produced by White Adipose Tissue Modulate Lipolysis in Lean but Not in Obese Rodent and Human

Chloe Buch et al.

Summary: The activation of the endocannabinoid system in white adipose tissue limits fat mobilization, potentially participating in progressive tissue remodeling leading to organ dysfunction. Acute CB1R blockade is ineffective in regulating lipolysis in obese WAT, suggesting a possible alteration of CB1R signaling in obesity conditions.

Functional Selectivity of a Biased Cannabinoid-1 Receptor (CB1R) Antagonist

Summary: The study found that the peripheral CB1R antagonist MRI-1891 is highly biased towards inhibiting CB1R-induced beta-arrestin-2 activity, reducing food intake and body weight without eliciting anxiety while improving muscle insulin resistance. In contrast, the global CB1R antagonist rimonabant may cause anxiety and does not involve beta-arrestin-2.
