Role of sphingolipid metabolism in neurodegeneration

Manal Alaamery et al.

Summary: Sphingolipid metabolism plays a crucial role in neurodegenerative diseases, affecting neuron development, health, and inducing neurotoxicity in the CNS. Gangliosides and sphingosine-1-phosphate contribute significantly to the pathogenesis of neurologic diseases and hold potential as therapeutic targets for the modulation of neurodegeneration.

Lipid metabolism in astrocytic structure and function

James A. K. Lee et al.

Summary: Astrocytes in the central nervous system play crucial roles in metabolic homeostasis, blood brain barrier regulation, and neuronal crosstalk; Lipids are fundamental for astrocyte function, including energy generation, membrane fluidity, and cell to cell signaling; Disruption in lipid metabolism, structure, and signaling in astrocytes can lead to pathogenic mechanisms associated with various neurological disorders.

The Protective Mechanism of Deuterated Linoleic Acid Involves the Activation of the Ca2+ Signaling System of Astrocytes in Ischemia In Vitro

Egor A. Turovsky et al.

Summary: Ischemia-like conditions followed by reoxygenation cause cell death in cerebral cortex cells due to induction of necrosis and apoptosis. D4-Lnn, a deuterated form of linoleic polyunsaturated fatty acid, inhibits necrosis and reduces apoptotic cell death by activating astrocytes through the phosphoinositide calcium system, leading to cytoprotection. The cytoprotective mechanism of D4-Lnn involves inhibition of OGD-induced calcium ions, increase in cytosolic and reactive oxygen species production, enhancement of protective gene expression, and suppression of damaging proteins.

Neurotoxic reactive astrocytes induce cell death via saturated lipids

Kevin A. Guttenplan et al.

Summary: Astrocytes respond to diseases and injuries in the central nervous system by releasing long-chain saturated fatty acids contained in lipoparticles, leading to the death of neurons and mature oligodendrocytes. The elimination of saturated lipids produced by astrocytes can mitigate astrocyte-induced toxicity, suggesting a mechanism by which astrocytes kill cells in the central nervous system.

Brain lipidomics as a rising field in neurodegenerative contexts: Perspectives with Machine Learning approaches

Daniel Baez Castellanos et al.

Summary: Lipids play essential roles in cellular functioning, particularly in neurodegenerative diseases, and their dysregulation has been linked to the etiology and progression of such diseases. The integration of lipidomics data with machine learning has shown promise in identifying new biomarkers and unveiling new molecular mechanisms associated with neurodegeneration.

Identification of Lipid Heterogeneity and Diversity in the Developing Human Brain

Aparna Bhaduri et al.

Summary: The lipidome, understudied but fundamental to life, was profiled in 154,910 single cells across different developmental ages and brain regions using single-cell mass spectrometry-based protocols, resulting in a unique lipid atlas of the developing human brain. Differentially expressed lipids were identified across brain structures, cortical areas, and developmental ages, enabling the inference of lipid profiles of major cell types and the detection of putative celltype specific lipids from the data set. This data set will facilitate further investigation of the developing human brain lipidome.
