Comparative Physical Study of Three Pharmaceutically Active Benzodiazepine Derivatives: Crystalline versus Amorphous State and Crystallization Tendency

Summary: This study investigates the strategies of chemical derivatization and amorphization for improving drug solubility and bioavailability, and explores how small differences in molecular structure of three related pharmaceutical compounds affect their crystalline structure, relaxation dynamics, and tendency towards recrystallization. The results show that the presence of N-H center dot center dot center dot O hydrogen bonds in both crystalline and amorphous phases significantly influences the characteristic temperatures of the compounds. Additionally, the study suggests that the kinetics of recrystallization of amorphous pharmaceuticals is not solely determined by structural or conformational relaxation dynamics, but also correlated with crystal growth rate and the presence of hydrogen bonding.
