Prognostic Biomarkers in Uveal Melanoma: The Status Quo, Recent Advances and Future Directions

Nuno Jorge Lamas et al.

Summary: Uveal melanoma is the most common cancer that develops inside adult eyes, with a poor prognosis and limited treatment options. There is a need to find methods that can accurately predict the prognosis of the disease and effective therapeutic strategies. This article summarizes the current knowledge and challenges in the field of uveal melanoma research, and discusses potential future directions.

The Interplay of Cx26, Cx32, Cx37, Cx40, Cx43, Cx45, and Panx1 in Inner-Ear Development of Yotari (dab1-/-) Mice and Humans

Josip Lesko et al.

Summary: By investigating the deficiency of DAB1-protein in the inner-ear development of yotari, we found that most connexins were significantly decreased in yotari, compared to wild-type mice and humans. Additionally, the expression of pannexin PANX1 was also significantly lower in yotari, except at certain developmental stages. These results emphasize the importance of connexins in the development of vestibular and cochlear functions and provide potential therapeutic targets for inner-ear impairments.

Mechanisms of Connexin Regulating Peptides

D. Ryan King et al.

Summary: Gap junctions (GJ) and connexins are crucial in cellular physiology and have been investigated for their potential therapeutic applications in various diseases. Modulating GJ signaling pathways and designing peptides have shown promise as attractive targets for therapeutic intervention. However, the overall mechanisms of action of connexin targeting peptides are still not fully understood despite years of research.

Differential Action of Connexin Hemichannel and Pannexin Channel Therapeutics for Potential Treatment of Retinal Diseases

Mohd N. Mat Nor et al.

Summary: This study investigated the effects of using the retinal therapeutic drugs probenecid and tonabersat in light-damaged rat retinas. Results showed that tonabersat alone was more effective than probenecid, and probenecid did not enhance the therapeutic effect of connexin hemichannel block in this model.

Pannexin 1 binds beta-catenin to modulate melanoma cell growth and metabolism

Samar Sayedyahossein et al.

Summary: The study revealed a direct interaction between PANX1 and beta-catenin to modulate growth and metabolism in melanoma cells, providing mechanistic insight into melanoma progression. This interaction may be a new component of the Wnt signaling pathway in various contexts.

An Update on Connexin Gap Junction and Hemichannels in Diabetic Retinopathy

Jorge Gonzalez-Casanova et al.

Summary: Diabetic retinopathy is a major cause of vision loss in the working age population, characterized by progressive deterioration of retinal microvasculature due to long-term metabolic changes. Gap junction channels and connexin hemichannels have been identified as key factors in the pathogenesis of diabetic retinopathy. Understanding the physiological impact of diabetes on these channels and their role in the disease process could lead to the development of novel pharmacological interventions.
