Tissue Niches Formed by Intestinal Mesenchymal Stromal Cells in Mucosal Homeostasis and Immunity

Col6a1+/CD201+ mesenchymal cells regulate intestinal morphogenesis and homeostasis

Summary: This study investigates the various subsets of intestinal mesenchymal cells and their roles in intestinal morphogenesis and pathophysiology. The results show that Col6a1(Cre) mouse targets fibroblast subsets and perivascular cells, as well as mesenchymal aggregates crucial for intestinal development. In adulthood, Col6a1(+)/CD201(+) mesenchymal cells regulate enteroendocrine cell differentiation and epithelial proliferation. During acute colitis, they exhibit an inflammatory and extracellular matrix remodelling gene signature. However, these cells are dispensable for normal organ architecture in both homeostasis and tissue regeneration.

This information suggests that innovative therapies validated by reliable clinical trial endpoints may prevent or reverse intestinal strictures.

Summary: NOD2 deficiency contributes to fibrosis and stricturing complications in Crohn's disease through dysregulated homeostasis of activated fibroblasts and macrophages, which can be ameliorated by gp130 blockade. Carriers of NOD2 risk alleles exhibit dysregulated homeostasis of activated fibroblasts and macrophages, suggesting a potential role for gp130 blockade in rescuing the activated program caused by NOD2 mutations.

Delineating proinflammatory microenvironmental signals by ex vivo modeling of the immature intestinal stroma

Summary: The disruption of intestinal stromal cells in pre-term infants may contribute to excessive inflammation, with upregulated TLR4 in the intestinal epithelium playing a key role. Through ex vivo co-culturing and genetic analysis, it was found that embryonic stromal cells promote the differentiation of monocytes into specific phagocytes, and that immature stromal cells exhibit different TLR signaling components compared to adult cells.

Induction of Il33 upon DSS exposure suggests that epithelial injury induces its expression, with fibroblasts being the main cellular source, and their expression increased during DSS-induced colitis.
