Inhibition of NETosis for treatment purposes: friend or foe?

Tahmineh Mohammadi Chamardani et al.

Summary: Active neutrophils release neutrophil extracellular traps (NETs), which play an important role in both innate and adaptive immune responses. In addition to defense against infections, NETosis is involved in the pathogenesis of several diseases. Targeting NET has become an important therapeutic approach.

Neutrophil extracellular traps and their histones promote Th17 cell differentiation directly via TLR2

Alicia S. Wilson et al.

Summary: This study reveals the mechanism by which neutrophil extracellular traps (NETs) directly activate T cells and specifically enhance Th17 cell differentiation. This suggests a direct link between neutrophils, NETs, and T cell autoimmunity.

Increased Circulating Cell-Free DNA in Eosinophilic Granulomatosis With Polyangiitis: Implications for Eosinophil Extracellular Traps and Immunothrombosis

Teppei Hashimoto et al.

Summary: In antineutrophil cytoplasmic antibody-associated vasculitis (AAV), levels of cfDNA are increased and associated with disease activity. Among AAV patients, those with eosinophilic granulomatosis with polyangiitis (EGPA) had the highest levels of cfDNA, and in vitro experiments showed that EETs are more stable against DNase and provide a scaffold for platelet adhesion.

Disulfiram inhibits neutrophil extracellular trap formation and protects rodents from acute lung injury and SARS-CoV-2

Jose M. Adrover et al.

Summary: Severe acute lung injury, characterized by high mortality and limited treatment options, can be improved with the use of disulfiram, an FDA-approved drug for alcohol use disorder. Disulfiram reduces neutrophil extracellular traps (NETs), improves survival and lung function in a mouse model of transfusion-related acute lung injury, and shows potential benefits in reducing NETs and inflammation in SARS-CoV-2-infected hamsters.

Multiple Origins of Extracellular DNA Traps

Edgar Ramos-Martinez et al.

Summary: Extracellular DNA traps (ETs) are evolutionarily conserved antimicrobial mechanisms found in protozoa, plants, and animals. By comparing similarities in ETs across different taxa, it is suggested that ETs may have multiple origins. The presence of a mechanism initiating the formation of ETs in multicellular organisms indicates a relationship between multicellularity and the development of ETs.

Role of Eosinophils in Intestinal Inflammation and Fibrosis in Inflammatory Bowel Disease: An Overlooked Villain?

Inge Jacobs et al.

Summary: Eosinophils, a type of white blood cell, play a crucial role in gastrointestinal health and are increasingly being studied for their inflammatory and pro-fibrotic functions in inflammatory bowel disease and other chronic inflammatory conditions. Research focus is shifting towards innate immunity and understanding the inflammatory properties of eosinophils.

Neutrophil extracellular traps promote gastric cancer metastasis by inducing epithelial-mesenchymal transition

Tong Zhu et al.

Summary: Neutrophil extracellular traps (NETs) in gastric cancer (GC) are associated with tumor progression, metastasis risk, and tumor microenvironment. The study found that NETs formation is affected by the tumor microenvironment and may exacerbate GC progression through promoting epithelial-mesenchymal transition (EMT). Hence, targeting NETs may be therapeutically beneficial in GC treatment.

Neutrophil Extracellular Traps: A Potential Therapeutic Target in MPO-ANCA Associated Vasculitis?

Kim M. O'Sullivan et al.

Summary: Understanding the role of cell death in autoimmune diseases can help define critical pathways in immunopathogenesis and identify new therapeutic targets. Neutrophils, crucial immune responder cells, play important roles in diseases like MPO-ANCA Microscopic Vasculitis in both host defense and autoimmunity.

Neutrophil Extracellular Traps Activate Proinflammatory Functions of Human Neutrophils

Daniel Doemer et al.

Summary: This study provides a comprehensive analysis of the effects of NETs on human primary neutrophils in vitro. The results demonstrate that NETs can activate various neutrophil functions, including ROS production, phagocytosis and killing of microbial pathogens. Additionally, NETs induce the secretion of inflammatory cytokines, further supporting their role in amplifying inflammatory events.

Phagocyte extracellular traps in children with neutrophilic airway inflammation

Paul T. King et al.

Summary: Childhood lung infections are often associated with neutrophilic airway inflammation and excess protease production. Extracellular traps by neutrophils and macrophages play a role in this inflammation. DNase 1 and AAT were found to have an impact on the process, and current therapeutics can regulate this innate inflammatory response in children.
