A Phase 1b Safety Study of SER-287, a Spore-Based Microbiome Therapeutic, for Active Mild to Moderate Ulcerative Colitis

Matthew R. Henn et al.

Summary: In this study, a phase 1b trial was conducted in 58 adults with active mild-to-moderate UC to evaluate the safety and efficacy of SER-287, an oral formulation of Firmicutes spores. The results showed that patients who received vancomycin preconditioning and daily doses of SER-287 had a higher clinical remission rate at week 8 compared to placebo, indicating that engraftment of dose species was facilitated by vancomycin preconditioning and daily dosing of SER-287.

Summary: The first-in-human study on intratumoral injection of Clostridium novyi-NT demonstrated its feasibility with manageable toxicities. The treatment induced antitumor responses and host immune reactions in patients, with most experiencing reduction or stability in injected tumor size. Further studies are warranted to explore the potential of C. novyi-NT in human therapy.

The Sporobiota of the Human Gut

Muireann Egan et al.

Summary: The human gut microbiome is a diverse ecosystem that is integral to human health. Studies have shown that the formation of endospores by specific members of the gut microbiome is linked to antibiotic resistance and the spread of resistance genes, known as the resistome. This has important implications for human health, disease, and biotechnology.

Host adaptation in gut Firmicutes is associated with sporulation loss and altered transmission cycle

Hilary P. Browne et al.

Summary: The human-to-human transmission of symbiotic anaerobic bacteria is crucial for the human gut microbiota. However, the genomic and biological adaptations involved in this transmission are still not well understood. The loss of sporulation in gut Firmicutes is associated with features of host-adaptation such as genome reduction and specialized metabolic capabilities. This suggests a trade-off between transmission range and colonization abundance in gut Firmicutes.
