Amyloid Oligomers: A Joint Experimental/Computational Perspective on Alzheimer's Disease, Parkinson's Disease, Type II Diabetes, and Amyotrophic Lateral Sclerosis

Phuong H. Nguyen et al.

Summary: Protein misfolding and aggregation are common in amyloidogenic diseases, and understanding how amyloid species become toxic is crucial for treatment development. Experiments using computer modeling, in vitro and in vivo studies, as well as pharmacology, have provided valuable insights into the accumulation and deposition of amyloid species, contributing to research on diseases like Alzheimer's, Parkinson's, diabetes, and ALS.

Nucleation-dependent Aggregation Kinetics of Yeast Sup35 Fragment GNNQQNY

Gunasekhar Burra et al.

Summary: This study used a reproducible solubilisation protocol to generate a homogenous monomeric solution of the yeast prion protein Sup35 N-terminal hepta-peptide sequence GNNQQNY, revealing molecular details of its self-assembly mechanism. The aggregation kinetics data showed that the GNNQQNY sequence follows nucleation-dependent aggregation kinetics with a critical nucleus of size similar to 7 monomers, where the efficiency of nucleation was inversely related to the reaction temperature.

A kinetic ensemble of the Alzheimer's Aβ peptide

Thomas Lohr et al.

Summary: The article discusses the conformational and thermodynamic properties of disordered proteins and proposes a method to describe the transition rates between different states using kinetic ensembles. By developing a Markov state model, the kinetic ensemble of A beta 42 is studied, revealing characteristics of state transitions on the microsecond timescale.

Summary: This study analyzed molecular dynamics simulations of Aβ peptide using different force fields to study its structure and dynamics, providing answers to important questions related to Aβ, such as suitable force fields for modeling, the actual appearance of Aβ peptide, and the time required for MD simulations to reach equilibrium. The analyses presented in this study will serve as a useful reference guide for understanding the behavior of Aβ peptide and other similar disordered proteins.
