Targeting Interleukin-10 Restores Graft Microvascular Supply and Airway Epithelium in Rejecting Allografts

Shadab Kazmi et al.

Summary: This study demonstrates that IL-10 can promote microvascular and epithelial repair in airway transplantation by regulating the expression of FOXJ1, beta-catenin, and Fascin-1 proteins. These findings provide an important therapeutic strategy for graft repair using IL-10.

Manipulating the Metabolism to Improve the Efficacy of CAR T-Cell Immunotherapy

Marsha Pellegrino et al.

Summary: The adoptive transfer of CAR-expressing T cells has shown unprecedented success in treating B-cell malignancies, but its efficacy in other types of cancers is still limited. The metabolic fitness of CAR T cells plays a crucial role in their ability to target and function within solid tumors, with both tumor and T cell metabolism impacting the immune response significantly.

Metabolic support of tumour-infiltrating regulatory T cells by lactic acid

McLane J. Watson et al.

Summary: T-reg cells exhibit broad heterogeneity in glucose metabolism within normal and transformed tissues, utilizing an alternative metabolic pathway to maintain their suppressive function and proliferation. Glucose uptake is correlated with poorer suppressive function and long-term instability, while high-glucose conditions impair T-reg cell function and stability. Additionally, T-reg cells upregulate pathways involved in the metabolism of lactic acid, with lactate uptake being dispensable for peripheral T-reg cells but required intratumorally for slowed tumor growth and increased response to immunotherapy.

T-cell Immunometabolism: Therapeutic Implications in Organ Transplantation

Danh T. Tran et al.

Summary: Despite significant progress in solid-organ transplantation over the past 110 years, there are still many unresolved issues that require advanced therapeutic strategies. The rapidly advancing field of T-cell immunometabolism has shown promise in developing novel therapeutic approaches, including for antitumor therapies, antimicrobial therapies, and autoimmune diseases. Further exploration of T-cell immunometabolism and potential therapeutic targets could lead to improved outcomes in solid-organ transplantation.

Integrating CAR T-Cell Therapy and Transplantation: Comparisons of Safety and Long-Term Efficacy of Allogeneic Hematopoietic Stem Cell Transplantation After CAR T-Cell or Chemotherapy-Based Complete Remission in B-Cell Acute Lymphoblastic Leukemia

Yan-Li Zhao et al.

Summary: In B-ALL patients, similar clinical safety outcomes could be achieved with either CD19 CAR T-cell therapy followed by allo-HSCT or chemotherapy followed by allo-HSCT. Despite the inclusion of more patients with advanced diseases in the CAR-T group, the 4-year LFS and OS achieved with CAR T-cells followed by allo-HSCT were as remarkable as those achieved with chemotherapy followed by allo-HSCT.

Butyrate enhances CPT1A activity to promote fatty acid oxidation and iTreg differentiation

Fengqi Hao et al.

Summary: The short-chain fatty acid butyrate not only promotes the differentiation of regulatory T (iTreg) cells, but also accelerates fatty acid oxidation to facilitate this process. Researchers found that by converting butyrate into butyryl-CoA (BCoA) through ACSS2, activity of CPT1A can be increased to promote iTreg differentiation.
