Structural Insights into Methylated DNA Recognition by the Methyl-CpG Binding Domain of MBD6 from Arabidopsis thaliana

Summary: This study determined the structure of the MBD domain of the MBD6 protein in Arabidopsis thaliana and investigated its binding properties to methylated DNA. The binding affinity of the MBD domain for methyl-CpG-containing DNA was found to be lower than that of mammalian MBD domains, due to the absence of positively charged residues. This research provides insights into how MBD proteins mediate epigenetic signals in plant cells.

MBD5 and MBD6 couple DNA methylation to gene silencing through the J-domain protein SILENZIO

Summary: The study identified two Arabidopsis thaliana methyl-CpG-binding domain proteins, MBD5 and MBD6, which are recruited to chromatin by recognition of CG methylation and redundantly repress a subset of genes and transposons without affecting DNA methylation levels. These methyl readers recruit a J-domain protein, SILENZIO, which functions as a transcriptional repressor downstream of DNA methylation. The conserved J-domain motif of SILENZIO is required for its interaction with HSP70s and its silencing function.
