Identification and Characterization of a Novel Thermostable GDSL-Type Lipase from Geobacillus thermocatenulatus

Summary: In this study, two putative genes encoding lipolytic enzymes from Geobacillus thermocatenulatus KCTC 3921 were cloned and overexpressed in Escherichia coli. The recombinant Lip29 and Est29 showed different specific activities and substrate specificities towards fatty acids of varying chain lengths. Lip29 exhibited thermostability and high tolerance against detergents and metal ions, with the highest binding affinity towards long-chain substrates. This study presents the first GDSL-type lipase identified in thermophilic bacteria, as revealed through biochemical and computational analyses.

Optimization, purification, and biochemical characterization of thermoalkaliphilic lipase from a novel Geobacillus stearothermophilus FMR12 for detergent formulations

The optimized fermentation parameters significantly increased the productivity of the TA lipase, which demonstrated outstanding activity and stability in extreme conditions. The purified enzyme showed high specific activity and fold purification, with potential for detergent formulations.

Structure elucidation and docking analysis of 5M mutant of T1 lipase Geobacillus zalihae

The three-dimensional structure analysis of the 5M mutant lipase revealed a decrease in molecular interactions, emphasizing the importance of hydrogen bonds and ion interactions for protein stability. Substrate specificity analysis showed changes in substrate preference, with molecular dynamics simulation validating the preference of 5M lipase towards long-chain p-nitrophenyl-esters.
