Transcriptional regulatory network for the establishment of CD8+ T cell exhaustion

Wooseok Seo et al.

Summary: Exhausted CD8(+) T cells may be deliberately generated and maintained to provide mild immune responses against chronic infection or cancer, which can be safer over the long term compared to strong immune responses. A specific population of exhausted CD8(+) T cells that behaves like self-renewing stem cells has been identified, suggesting they can be considered progenitors of exhausted CD8(+) T cells. Studies have revealed the regulatory network governing the generation and propagation of exhausted CD8(+) T cells, shedding light on the developmental stages that lead to this subtle but effective immune response.

Inhibition of BCL11B induces downregulation of PTK7 and results in growth retardation and apoptosis in T-cell acute lymphoblastic leukemia

Kehan Li et al.

Summary: The study identified PTK7 as a potential downstream target of BCL11B in T-ALL. Inhibition of the BCL11B pathway leads to reduced PTK7 expression, which induces growth retardation and apoptosis in T-ALL cells via up-regulation of TRAIL and p27.

The role of NFAT2/miR-20a-5p signaling pathway in the regulation of CD8+ naive T cells activation and differentiation

Yikai Zhang et al.

Summary: T cell dysfunction is a common characteristic in leukemia patients, with lower expression of NFAT2 and pri-miR-17 similar to 92 found in CD3(+) T cells from CML patients. The regulation of CD8(+) T cell function by NFAT2 and miR-20a is crucial, suggesting that they may be potential targets for revising T cell function in leukemia immunotherapy.

NFAT transcription factors are essential and redundant actors for leukemia initiating potential in T-cell acute lymphoblastic leukemia

Claire Catherinet et al.

Summary: NFAT transcription factors play crucial roles in T-ALL, essential for the development of leukemia cells. Nfat genes show functional redundancy in T-ALL and may serve as future candidate targets for therapy.

M2 macrophages, but not M1 macrophages, support megakaryopoiesis by upregulating PI3K-AKT pathway activity

Hong-Yan Zhao et al.

Summary: The study revealed the critical role of macrophages in regulating megakaryocyte production, with M1 and M2 macrophages exhibiting opposing effects in a manner dependent on the PI3K-AKT pathway. This sheds light on the pathogenesis of thrombocytopenia and presents a potential therapeutic strategy for promoting megakaryopoiesis.
